Sulfatase 1 also known as SULF1 or HSulf-1 is an enzyme that functions by removing sulfate groups from heparan sulfate proteoglycans. Its molecular mass is approximately 125 kDa. SULF1 is mainly expressed in the liver and it is also found in the kidney thyroid and skin tissues. It possesses a sulfatase activity that acts on the 6-O-position of heparan sulfate thereby modulating the binding sites for several growth factors.
Biological function summary

SULF1 plays a significant role in modulating cellular processes like growth angiogenesis and tissue repair by altering the bioavailability of heparan sulfate-bound signaling molecules. It often acts as a monomer but may participate in larger complexes during functional modulation. SULF1 regulates interactions with key growth factors such as fibroblast growth factors (FGFs) and vascular endothelial growth factors (VEGFs) impacting cell communication and migration.

Pathways

SULF1 is an important regulator in signaling pathways including the VEGF signaling pathway and the FGF signaling pathway. These pathways are essential for processes like angiogenesis and embryonic development. SULF1 activity influences these pathways by modifying heparan sulfate chains affecting the binding of FGFs and VEGFs to their respective receptors alongside related proteins like SULF2 which shares similar functions in these pathways.

SULF1's modulation of cellular signaling makes it relevant in cancer and fibrosis where aberrant signaling contributes to disease progression. Altered SULF1 expression has been associated with certain carcinomas such as hepatocellular carcinoma where it can influence tumor growth and metastasis. It can remove sulfate from the C-6 position of glucosamine within specific subregions of intact heparin (PubMed : 12368295, PubMed : 12686563). Diminishes HSPG (heparan sulfate proteoglycans) sulfation, inhibits signaling by heparin-dependent growth factors, diminishes proliferation, and facilitates apoptosis in response to exogenous stimulation (PubMed : 12686563).
