Sulfatase 2 (SULF2) also known as SULF-2 or extracellular sulfatase acts as an enzyme that removes sulfate groups from the heparan sulfate proteoglycans (HSPGs). It is a 100-110 kDa protein located primarily on the cell surface. SULF2's expression occurs mainly in tissues such as the liver kidney and lungs although it can be found in various other tissues too. By modifying HSPGs SULF2 influences cell signaling events that are critical to cellular communication and function.
Biological function summary

SULF2 participates in the modulation of growth factor signaling and cell surface interactions. This enzyme does not exist in isolation; instead it functions as a part of complexes involved in the extracellular matrix (ECM). Through its catalytic action SULF2 modulates the availability and activity of signaling molecules such as fibroblast growth factors (FGFs) and Wnt proteins. It impacts processes such as cell proliferation differentiation and developmental processes by altering the structural properties of the ECM.

Pathways

SULF2 significantly modulates the Wnt signaling pathway and the fibroblast growth factor receptor (FGFR) pathway. These pathways play essential roles in development and tissue regeneration. By altering the sulfation states of heparan sulfate chains SULF2 affects the interaction of proteins like Frizzled receptors in the Wnt pathway and FGFRs in the FGF signaling pathway. This modulation leads to changes in signal transduction further influencing the cellular responses during growth and development.

SULF2 has been linked with several cancer types and neurodegenerative diseases. In cancer such as hepatocellular carcinoma SULF2 overexpression promotes tumor growth and invasion by influencing growth factor signaling. It often associates with proteins like β-catenin in cancer-related pathways. In neurodegenerative diseases SULF2 modulates neural stem cell differentiation and repair influencing conditions like Alzheimer's disease. The enzyme's interaction with amyloid-beta peptides and tau protein can impact disease progression and severity.
