Function

Sulfotransferase that utilizes 3'-phospho-5'-adenylyl sulfate (PAPS) as sulfonate donor to catalyze the sulfate conjugation. Responsible for the sulfation of cholesterol (PubMed:12145317, PubMed:19589875). Catalyzes sulfation of the 3beta-hydroxyl groups of steroids, such as, pregnenolone and dehydroepiandrosterone (DHEA) (PubMed:12145317, PubMed:16855051, PubMed:21855633, PubMed:9799594). Preferentially sulfonates cholesterol, while it has also significant activity with pregnenolone and DHEA (PubMed:12145317, PubMed:21855633). Plays a role in epidermal cholesterol metabolism and in the regulation of epidermal proliferation and differentiation (PubMed:28575648). Isoform 2. Sulfonates pregnenolone but not cholesterol.

Activity summary

SULT2B1 also known as sulfotransferase family 2B member 1 functions as a cytosolic enzyme that adds sulfate groups to various molecules. This post-translational modification is important for the metabolism of hormones neurotransmitters drugs and xenobiotic compounds. SULT2B1 has a molecular mass of approximately 42 kDa. It is expressed in a variety of tissues with higher levels in the skin brain and reproductive organs indicating its diverse physiological roles across different systems in the body.

Biological function summary

The role of SULT2B1 involves the sulfation of cholesterol and dehydroepiandrosterone (DHEA) which are important for maintaining lipid homeostasis and steroid hormone biosynthesis. The enzyme is not part of a complex but works optimally in its standalone cytosolic environment. It participates actively in regulating the availability of active hormones and lipids making it a significant player in cellular metabolic processes.

Pathways

SULT2B1 operates within two main biological frameworks: the steroid hormone biosynthesis pathway and lipid metabolic pathway. In these pathways the enzyme interacts closely with cholesterol and DHEA to modulate their sulfation status influencing precursor availability for steroidogenesis. It works in conjunction with other sulfotransferases like SULT1A1 that also help manage the sulfate pool important for hormone regulation and metabolic balance.

Associated diseases and disorders

Aberrations in SULT2B1 function can link to conditions like polycystic ovary syndrome (PCOS) and certain cancers due to its role in steroid hormone metabolism. Disruptions in the enzyme's activity could alter DHEA and cholesterol sulfation leading to hormone imbalances that contribute to these disorders. In the context of PCOS associated enzymes might include subtypes of cytochrome P450 which function alongside SULT2B1 in steroid biosynthesis pathways to maintain hormone equilibrium.
