As a component of the LINC (LInker of Nucleoskeleton and Cytoskeleton) complex involved in the connection between the nuclear lamina and the cytoskeleton (PubMed:18039933, PubMed:18396275). The nucleocytoplasmic interactions established by the LINC complex play an important role in the transmission of mechanical forces across the nuclear envelope and in nuclear movement and positioning (By similarity). Required for interkinetic nuclear migration (INM) and essential for nucleokinesis and centrosome-nucleus coupling during radial neuronal migration in the cerebral cortex and during glial migration (By similarity). Involved in telomere attachment to nuclear envelope in the prophase of meiosis implicating a SUN1/2:KASH5 LINC complex in which SUN1 and SUN2 seem to act at least partial redundantly (By similarity). Required for gametogenesis and involved in selective gene expression of coding and non-coding RNAs needed for gametogenesis (By similarity). Helps to define the distribution of nuclear pore complexes (NPCs) (By similarity). Required for efficient localization of SYNE4 in the nuclear envelope (By similarity). May be involved in nuclear remodeling during sperm head formation in spermatogenesis (By similarity). May play a role in DNA repair by suppressing non-homologous end joining repair to facilitate the repair of DNA cross-links (PubMed:24375709).

SUN1 also known as Sad1-Unc-84 homology 1 is a protein with a molecular mass of approximately 97 kDa. This protein is an important component of the nuclear envelope which helps maintain the structure of the nucleus. It is expressed in various tissues including muscle and neurons and is an integral part of the linking complex that connects the nuclear envelope to the cytoskeleton. SUN1 functions as a structural protein and contributes to the mechanical stability of the nuclear architecture.
SUN1 plays significant roles in cellular functions through its interaction with other proteins. It is an essential part of the LINC (Linker of Nucleoskeleton and Cytoskeleton) complex where it interacts with KASH domain-containing proteins that span the outer nuclear membrane. This interaction is important for nuclear positioning centrosome attachment and chromosomal movement. SUN1 participates in critical processes such as DNA repair and meiotic progression influencing cell cycle regulation and genetic stability.
SUN1 integrates into cellular communication systems by mediating mechanical signals across the nuclear-cytoplasmic boundary. It is actively involved in the nuclear envelope-endoplasmic reticulum network which is critical for nuclear-cytoskeletal coupling. The protein associates with SUN2 another member of the SUN family and they both interact with the Nesprin proteins anchoring the nucleus to the cell's structural framework. This association enables SUN1 to influence signaling pathways related to cell division and differentiation.
Mutations and malfunctions within the SUN1 protein have been associated with several medical conditions. Its role is particularly noted in certain muscular dystrophies and nuclear envelopathies where mutations disrupt nuclear-cytoskeletal connections. In these pathological states SUN1 has links with proteins like Emerin which also reside at the nuclear envelope and whose dysfunction causes Emery-Dreifuss muscular dystrophy. These interconnections highlight the importance of SUN1 in maintaining cellular integrity and function particularly in muscle and nerve cells.

Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) analysis of human breast tissue sections labeling SUN1 with purified ab124770 at 1/200 dilution (0.698 µg/ml). Perform heat mediated antigen retrieval using Citrate buffer, pH 6.0. ImmunoHistoProbe one step HRP Polymer (ready to use) was used as the secondary antibody. Negative control: PBS instead of the primary antibody. Hematoxylin was used as a counterstain.
The bands observed are consistent with what has been described in PMID: 16648470
