As a probable component of the LINC (LInker of Nucleoskeleton and Cytoskeleton) complex, involved in the connection between the nuclear lamina and the cytoskeleton. The nucleocytoplasmic interactions established by the LINC complex play an important role in the transmission of mechanical forces across the nuclear envelope and in nuclear movement and positioning. May be involved in nuclear remodeling during sperm head formation in spermatogenesis. A probable SUN3:SYNE1 LINC complex may tether spermatid nuclei to posterior cytoskeletal structures such as the manchette.

SUN3 also known as Sad1 and UNC-like domain-containing protein 3 has a molecular mass of approximately 115 kDa. This protein is mechanistically involved in nuclear envelope organization and is predominantly expressed in testicular tissue specifically within spermatogenic cells. It localizes to the nuclear envelope where it is believed to contribute to the structural integrity and function of the nuclear membrane.
SUN3 is involved in the formation of the LINC complex which stands for LInker of NucleOcytoskeleton Complex. This complex serves as a bridge between the nucleoskeleton and the cytoskeleton. Through this connection SUN3 assists in maintaining nuclear positioning and stability playing a role in the mechanical coupling of the nucleus with cytoskeletal elements. This interaction is essential for processes like spermatogenesis where nuclear movement and positioning are key.
SUN3 participates in the spermatogenesis and meiotic cell division processes. The LINC complex including SUN3 partners with proteins such as KASH5 in these pathways to ensure proper chromosomal movement and alignment. Its role in these pathways is critical for normal fertility as errors can lead to abnormal sperm development and function.
Alterations in the function or expression of SUN3 affect male fertility leading to conditions such as oligozoospermia or asthenozoospermia. These conditions arise when the structural and positional integrity of the sperm nucleus is compromised. SUN3 interactions with proteins like KASH5 are significant because any disruption can result in impaired spermatogenesis contributing to these fertility issues.
