Superoxide Dismutase 3 also known as EC-SOD is an antioxidant enzyme with a molecular weight of approximately 135 kDa. It's an extracellular enzyme that catalyzes the dismutation of superoxide radicals into hydrogen peroxide and molecular oxygen. EC-SOD is found mostly in the extracellular matrix tissues such as lungs blood vessels and heart. Its activity depends on the presence of metal ions copper and zinc leading its classification as a Cu/Zn SOD enzyme. Scientists often study EC-SOD using ELISA assays because of its importance in neutralizing reactive oxygen species.
The enzyme acts as an important regulator of oxidative stress by reducing levels of superoxide radicals. EC-SOD forms a complex with heparan sulfate proteoglycans in the extracellular matrix which influences its stability and distribution. This enzyme plays a role in maintaining the redox balance influencing cell signaling inflammation and tissue damage which makes it important in managing oxidative stress in different tissues including the cardiovascular and respiratory systems.
Superoxide dismutase 3 participates in antioxidant pathways important for cellular defense against oxidative stress. It is particularly important in the oxidative stress response pathway. By reducing superoxide radicals EC-SOD influences pathways involving other antioxidant proteins like catalase and glutathione peroxidase. These interactions help prevent the accumulation of harmful oxidants and maintain cellular homeostasis. EC-SOD's action connects closely with other superoxide dismutases such as Cu/Zn SOD and MnSOD within these protective pathways.
Enhanced or impaired EC-SOD function associates with various health issues such as cardiovascular diseases and chronic obstructive pulmonary disease (COPD). EC-SOD has been shown to protect vascular health by reducing oxidative stress related damages which can mitigate the risk of atherosclerosis. In the context of respiratory conditions like COPD alterations in EC-SOD expression can disrupt lung tissue function and inflammatory responses. Its interactions with proteins like catalase further contribute to its protective role against cell and tissue damage during disease progression.

Overlay histogram showing HEK293 cells stained with ab80946 (red line). The cells were fixed with 80% methanol (5 min) and incubated in 1x PBS / 10% normal goat serum / 0.3M glycine to block non-specific protein-protein interactions. The cells were then incubated with the antibody (ab80946, 2µg/1x106 cells) for 30 min at 22ºC. The secondary antibody used was DyLight® 488 goat anti-mouse IgG (H+L) (Goat Anti-Mouse IgG H&L (DyLight® 488) preadsorbed ab96879) at 1/500 dilution for 30 min at 22ºC. Isotype control antibody (black line) was mouse IgG1 [ICIGG1] (Mouse IgG1, Kappa Monoclonal [B11/6] - Isotype Control ab91353, 2µg/1x106 cells) used under the same conditions. Acquisition of >5,000 events was performed.

ab80946 at 1/100 dilution staining Superoxide Dismutase 3/EC-SOD in human cartilage by Immunohistochemistry (Bouin's fixative fixed paraffin-embedded tissue section). Antigen retrieval was done by microwave in citrate buffer. A HRP-conjugated anti mouse secondary was used at 1/10 dilution. Image 'a' repersents control, 'b' staining of tissue from young human cartilage and 'c' show staining cartilage of a person suffered from osteoarthritis.
