Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) - Anti-Surfactant Protein A/PSAP antibody (AB115791)

Surfactant Protein A/PSAP was detected in paraffin-embedded section of rat lung tissues. Heat mediated antigen retrieval was performed in citrate buffer (pH6, epitope retrieval solution) for 20 mins. The tissue section was blocked with 10% goat serum. The tissue section was then incubated with 1μg/ml ab115791 overnight at 4°C. Biotinylated goat antirabbit IgG was used as secondary antibody and incubated for 30 minutes at 37°C. The tissue section was developed using Strepavidin-Biotin-Complex with DAB as the chromogen.

Supplementary information

Surfactant Protein A also known as PSAP is an important component found in the lungs. It is a protein approximately 35-40 kDa in size. Surfactant Protein A is predominantly expressed in the alveolar type II cells and Clara cells within the respiratory tract. This protein plays a significant role in lung function helping to maintain the surface tension of the alveoli to prevent collapse and to assist in host defense mechanisms as part of the lung surfactant system.
Biological function summary

Surfactant Protein A contributes to processes involving immune defense and inflammation control. It forms part of a larger surfactant complex and its main function is to bind and clear pathogens and allergens from the pulmonary system. The protein thereby plays an important role in recognizing foreign bodies promoting phagocytosis by macrophages and modulating inflammatory responses which is important for maintaining lung health and preventing infections.

Pathways

Surfactant Protein A interacts within immune signaling networks notably the lectin and complement activation pathways. These pathways facilitate the identification and clearance of pathogens. The protein stands in close relation to other members of the surfactant protein family such as Surfactant Protein D enhancing its functional scope in pulmonary immunity and host defense.

The malfunction or altered expression of Surfactant Protein A associates with respiratory conditions including pulmonary fibrosis and respiratory distress syndrome. Deficiencies or anomalies in this protein can also impact immune responses contributing to chronic obstructive pulmonary disease (COPD) by interacting with related proteins like Surfactant Protein B which is also important in surfactant function and maintenance of alveolar stability. Understanding these connections can aid in developing treatments targeting these pathways and disease mechanisms.

Target data

In presence of calcium ions, it binds to surfactant phospholipids and contributes to lower the surface tension at the air-liquid interface in the alveoli of the mammalian lung and is essential for normal respiration. Enhances the expression of MYO18A/SP-R210 on alveolar macrophages (PubMed : 25965346).
