Supplementary information

This supplementary information is collated from multiple sources and compiled automatically.

SUV3L1 also known as Suppressor of Var1 3-Like 1 is an ATP-dependent RNA helicase with a mass of approximately 90 kDa. Mechanically it unwinds RNA and participates in RNA processing and surveillance ensuring the integrity of RNA molecules. SUV3L1 exhibits a wide expression pattern being found in various tissues predominantly in regions with high mitochondrial activity due to its critical role in RNA metabolism.
Biological function summary

The helicase activity of SUV3L1 plays an important role in mitochondrial RNA degradation. SUV3L1 forms a functional complex with polynucleotide phosphorylase (PNPase) and together they contribute to mitochondrial RNA processing and decay. This complex is vital for maintaining mitochondrial RNA homeostasis and ensuring proper mitochondrial function affecting mitochondrial biogenesis and energy production.

Pathways

It is involved in mitochondrial gene expression and respiratory chain assembly. In the context of regulated RNA decay pathways SUV3L1 acts alongside PNPase to maintain proper levels of mitochondrial transcripts impacting cellular energy balance. Furthermore SUV3L1 is connected to the mitochondrial DNA transcription pathway where it associates with mitochondrial transcription factor B2 (TFB2M) enhancing the stability of mitochondrial transcripts and ensuring the effective production of mitochondrial proteins.

SUV3L1 has been implicated in mitochondrial disorders and certain types of cancer. Alterations in its function can lead to disrupted mitochondrial biogenesis contributing to disorders characterized by impaired mitochondrial activity. Research shows that SUV3L1 interacts with proteins like POLRMT which is part of the mitochondrial transcription machinery linking its dysregulation to mitochondrial-related dysfunctions and tumorigenesis. Understanding these associations helps illuminate SUV3L1's role in disease pathogenesis and potential therapeutic strategies.

Target data

Major helicase player in mitochondrial RNA metabolism. Component of the mitochondrial degradosome (mtEXO) complex, that degrades 3' overhang double-stranded RNA with a 3'-to-5' directionality in an ATP-dependent manner. Involved in the degradation of non-coding mitochondrial transcripts (MT-ncRNA) and tRNA-like molecules (PubMed : 29967381). ATPase and ATP-dependent multisubstrate helicase, able to unwind double-stranded (ds) DNA and RNA, and RNA/DNA heteroduplexes in the 5'-to-3' direction. Plays a role in the RNA surveillance system in mitochondria; regulates the stability of mature mRNAs, the removal of aberrantly formed mRNAs and the rapid degradation of non coding processing intermediates. Also implicated in recombination and chromatin maintenance pathways. May protect cells from apoptosis. Associates with mitochondrial DNA.
