Immunocytochemistry/ Immunofluorescence - Anti-Syk (phospho T317 + T323) Antibody [RP23040187] (AB313476)
Immunofluorscent analysis of Ramos cells fixed with 4% formaldehyde reconstituted in 1X PBS for 10 min at room temperature and permabilized using 0.1 % Triton X-100 in PBS for 15 min at room temperature labeling Syk pY317 pY323 with ab313476 at 2 µg/mL dilution followed by Goat anti-Rabbit IgG (H+L) Secondary Antibody, Alexa Fluor488 conjugate at 1/2000 dilution. Panel a) shows representative cells that were stained for detection and localization of Syk pY317 pY323 protein (green), Panel b) is stained for nuclei (blue) using DAPI. Panel c) represents cytoskeletal F-actin staining using Alexa Fluor 555 Rhodamine Phalloidin at 1/300 dilution. Panel d) is a composite image of Panels a, b and c clearly demonstrating membranous localization of Syk pY317 pY323. Panel e) shows loss of signal by competition with the Syk pY317 pY323 phospho peptide, demonstrating antibody specificity, and panel f) demonstrates no competition with the non-phosphorylated peptide. Panel g) represents control cells with no primary antibody to assess background. The images were captured at 60X magnification.

It is a part of signal transduction complexes and functions in the regulation of immune cell activation differentiation and survival. It affects processes like Fc receptor signaling in macrophages and neutrophils impacting immune responses and inflammation. The phospho-Syk form influences the strength and duration of signaling affecting various cellular functions.
Pathways
Syk is significant in the B-cell receptor (BCR) signaling pathway and Fc receptor signaling pathway. Within these pathways Syk interacts with proteins like Lyn and Fyn which are other kinases involved in BCR signal transduction. Syk phosphorylation noted as p-Syk plays a part in activating downstream molecules such as phospholipase C gamma (PLCγ) to mediate cellular responses to external stimuli.
