Supplementary information

Symplekin is a multifunctional protein essential for mRNA processing. It has a molecular mass of approximately 150 kDa and is alternatively known as SYM or SPK. You can find Symplekin expressed in the nucleus of eukaryotic cells where it acts mainly at the 3' end of mRNAs. This protein interacts with other molecules in the cell through its well-defined domains that facilitate its function in RNA processing.
Biological function summary

Symplekin modulates the stability and maturation of mRNA transcripts. It acts as a scaffold protein assembling components of the cleavage and polyadenylation specificity factor (CPSF) complex. Within this complex Symplekin collaborates with other proteins like CPSF160 and CPSF73 to regulate the addition of the poly(A) tail important for mRNA stability transport and translation. This function supports the processing of pre-mRNA into mature mRNA.

Pathways

Symplekin participates in important gene expression regulation. It plays a role in the mRNA processing and gene expression pathways. Symplekin interacts with proteins involved in these pathways like CPSF and other RNA-binding proteins ensuring accurate mRNA maturation and stability. These coordinated activities are vital for the regulation of gene expression and by extension cell function and viability.

Symplekin shows association with conditions related to abnormal mRNA processing. Dysregulation of Symplekin can have impacts on cellular processes and has been implicated in cancer and neurodegenerative diseases. In cancer improper mRNA processing and polyadenylation can lead to abnormal cell proliferation and tumor growth with Symplekin often interacting with oncogenic regulatory proteins. In neurodegenerative diseases such as Alzheimer's disruptions involving Symplekin might affect neural mRNA stability linking it to amyloid precursor protein processing.

Target data

Scaffold protein that functions as a component of a multimolecular complex involved in histone mRNA 3'-end processing. Specific component of the tight junction (TJ) plaque, but might not be an exclusively junctional component. May have a house-keeping rule. Is involved in pre-mRNA polyadenylation. Enhances SSU72 phosphatase activity.
