Activity summary

Syntaxin 18 also known as STX18 is a membrane protein that plays an important role in intracellular protein transport. This protein has an approximate molecular mass of 34.5 kDa and primarily resides in the endoplasmic reticulum (ER). Syntaxin 18 helps mediate vesicle trafficking facilitating the movement of different molecules between the ER and the Golgi apparatus. It acts as a target-specific SNARE protein which stands for "soluble N-ethylmaleimide-sensitive factor attachment protein receptor." SNARE proteins are known for their role in fusion processes of transport vesicles to their target membranes.

Biological function summary

Syntaxin 18 is involved in maintaining cellular homeostasis. It functions as part of a larger complex often referred to as the Syntaxin 18 complex which also includes other proteins such as SNARE partners Use1 BNIP1 and Sec22b. This protein complex is essential in the post-translational transport of proteins from the ER to the Golgi. Syntaxin 18 ensures precise vesicle targeting and fusion which is important for proper cell function and protein maturation processes within the secretory pathway.

Pathways

Syntaxin 18 integrates into the secretory pathway where it coordinates with other proteins like VAMP7 and SNAP29 to facilitate vesicle transport. This pathway is vital for the regulation of protein and lipid shipping within cells. Syntaxin 18 also plays an integral role in the ER-to-Golgi transport vesicles which are important in protein secretion and autophagy. As Syntaxin 18 is linked to proteins involved in vesicular trafficking it is critical in ensuring that cellular components are distributed efficiently within the cell.

Associated diseases and disorders

Syntaxin 18 has associations with neurodegenerative disorders such as Alzheimer's disease due to its role in protein traffic dysregulation. Abnormal expression or mutations in Syntaxin 18 can disrupt vesicular transport leading to cellular stress and pathological aggregates that relate to these diseases. Furthermore Syntaxin 18 interacts with proteins like VAMP7 which also impacts processes implicated in certain autoimmune diseases. Understanding these interactions provides insights into how impaired Syntaxin 18 function contributes to disease progression.
