Supplementary information

Syntaxin 7 also known as STX7 is a member of the SNARE protein family important for vesicle fusion processes. With a molecular mass of approximately 29 kDa Syntaxin 7 plays a significant role in the intracellular trafficking system by mediating membrane fusion events. It is expressed predominantly in many tissues including the brain and spleen where it contributes to diverse cellular functions. As a member of the target membrane SNARE proteins Syntaxin 7 partners with other SNARE proteins to facilitate endolysosomal transport.
Biological function summary

Syntaxin 7 mediates membrane fusion events essential for endosome-lysosome fusion. It forms a complex with other SNARE proteins like Vti1b Syntaxin 8 and VAMP8. This complex orchestrates the accurate trafficking and degradation of macromolecules within cells. Syntaxin 7's presence proves essential in ensuring molecules are delivered to lysosomes for degradation and it participates in recycling cellular components. This role helps maintain cellular homeostasis by regulating the turnover of cellular substances.

Pathways

Syntaxin 7 plays vital roles in processes related to vesicle transport and autophagy. It contributes significantly to the endocytic pathway an important pathway responsible for importing molecules into cells and directing them to lysosomes for breakdown. Syntaxin 7 is also involved in the autophagic-lysosomal pathway working in conjunction with proteins like Syntaxin 8 to facilitate autophagosome and lysosome fusion. Its coordination in these pathways highlights the importance of Syntaxin 7’s role in maintaining cellular health and balance.

Syntaxin 7 has been linked to conditions related to defective lysosomal storage and neurodegenerative diseases. In Alzheimer’s disease for instance impaired endosomal and lysosomal functions are typical and dysfunction of proteins like Syntaxin 7 can exacerbate these issues. Similarly abnormal expression of Syntaxin 7 could contribute to neuronal loss observed in Alzheimer's disease. Its interaction with other SNARE proteins in this context suggests that targeting Syntaxin 7 and its complex could offer therapeutic potential for addressing related cellular dysfunctions in these disorders.
