Flow Cytometry - Anti-TACI antibody [EPR23045-153] (AB239370)
Flow cytometric analysis of human peripheral blood mononuclear cell (PBMC) cells labeling TACI with ab239370 at 1/400 dilution (Right) compared with a Rabbit monoclonal IgG (ab172730) isotype control (Left). Goat anti rabbit IgG (Alexa Fluor® 488, ab150077) at 1/2000 dilution was used as the secondary antibody.
Cells were stained with rabbit IgG (Left) or ab239370 (Right). Then stained with anti-CD19 conjugated to Alexa Fluor 647.
Gated on viable cells.

Immunoprecipitation - Anti-TACI antibody [EPR23045-153] (AB239370)
TACI was immunoprecipitated from 0.35 mg Raji (Human Burkitt's lymphoma B lymphocyte) whole cell lysate 10μg with ab239370 at 1/30 dilution (2μg in 0.35mg lysates). Western blot was performed on the immunoprecipitate using ab239370 at 1/1000 dilution. VeriBlot for IP Detection Reagent (HRP) (ab131366) was used at 1/1000 dilution.

Lane 2 : ab239370 IP in Raji whole cell lysate
Lane 3 : Rabbit monoclonal IgG (ab172730) instead of ab239370 in Raji whole cell lysate

Biological function summary
TACI involves the regulation of B cell function and survival. It binds to two ligands APRIL and BAFF and TACI is not part of any larger complex. Upon ligand binding TACI can promote antibody class-switching which adjusts the type of antibody that B cells produce. This action supports the immune response by allowing the body to adapt to different pathogens effectively.
Pathways
Immune responses involve TACI in the BAFF-APRIL signaling pathway. The interplay between TACI and its ligands influences the NF-kB signaling cascade leading to B cell proliferation and survival. TACI interacts with other proteins such as BCMA and BAFF-R which are important in regulating B cell homeostasis and function ensuring balanced and effective immune responses.
