TAF1B also known as TATA box binding protein-associated factor RNA polymerase I subunit B is a component of the transcription machinery. It has a molecular weight of approximately 63 kDa. TAF1B plays an important role in the formation of the transcription initiation complex for RNA polymerase I. Its expression occurs mainly in the nucleolus of eukaryotic cells where it directly interacts with other transcription factors.
Biological function summary

TAF1B regulates the initiation of rRNA transcription. It forms part of the SL1 complex which is essential for recognizing the rDNA promoter and facilitating the binding of RNA polymerase I. The SL1 complex which includes TAF1B TBP TAF1A and TAF1C ensures the accuracy of rRNA gene transcription. This assembly directly influences ribosomal RNA (rRNA) synthesis a critical process in ribosome biogenesis in cells.

Pathways

TAF1B plays a major role in the ribosome biogenesis and cell growth pathways. Its function within the SL1 complex helps initiate the transcription of rRNA genes by RNA polymerase I. TAF1B collaborates with proteins such as TAF1A and TAF1C in these pathways. These pathways are fundamental for cell proliferation and protein synthesis emphasizing the importance of TAF1B in maintaining cellular function.

TAF1B associates with conditions such as cancer and ribosomopathies. Dysregulation of rRNA synthesis due to anomalies in TAF1B can lead to uncontrolled cellular growth a hallmark of cancer. Alterations in TAF1B's function have also been linked to ribosomal biogenesis disorders.
