Component of the transcription factor SL1/TIF-IB complex, which is involved in the assembly of the PIC (pre-initiation complex) during RNA polymerase I-dependent transcription. The rate of PIC formation probably is primarily dependent on the rate of association of SL1/TIF-IB with the rDNA promoter. SL1/TIF-IB is involved in stabilization of nucleolar transcription factor 1/UBTF on rDNA. Formation of SL1/TIF-IB excludes the association of TBP with TFIID subunits. Recruits RNA polymerase I to the rRNA gene promoter via interaction with RRN3.

TAF1C also known as Transcription Initiation Factor TFIID Subunit 1C is a component of the transcription factor complex that plays an important role in the initiation of transcription by RNA polymerase I. It has a molecular mass of approximately 64 kDa. TAF1C is mainly expressed in the nucleolus of eukaryotic cells where it aids in forming the transcription preinitiation complex. The protein interacts specifically with other subunits to enable the RNA polymerase I to initiate the transcription of ribosomal RNA genes.
TAF1C is essential for assembling the RNA polymerase I complex often referred to as the SL1 complex. This complex includes other components like TBP and several TAFs which together promote the transcription of rRNA genes critical for ribosome biogenesis. By participating in the formation of this complex TAF1C impacts cellular growth by influencing ribosome production. The functional activity of TAF1C thereby strongly ties to cellular metabolism given its position in rRNA transcription machinery.
TAF1C is involved in the rRNA synthesis pathway a pathway important for ribosome production. This pathway interfaces with the cell growth and division pathways with proteins like TBP and other transcription factors forming significant parts of these linked networks. TAF1C contributes to maintaining cellular homeostasis by its functions in these pathways impacting how cells respond and adapt to metabolic demands and growth signals.
TAF1C has connections to diseases like cancer due to its involvement in ribosome biogenesis. Altered expression of TAF1C affects nucleolar activity which is often observed in tumor cells. Moreover any disruption in its function can lead to dysregulation of rRNA synthesis linking it indirectly to p53 a protein involved in cell cycle regulation and apoptosis. Aberrant regulation or mutations in TAF1C-related pathways may fuel disease progression and are subjects of current research interest in oncology.
