TAF5L also known as Transcription initiation factor TFIID subunit 5-like is a protein that plays an important role in gene expression through its involvement in the transcription machinery. It weighs approximately 80 kDa and is composed of 787 amino acids. TAF5L is expressed in various tissues with higher levels in the spleen thymus and bone marrow. This expression pattern indicates its role in processes that require precise transcription regulation.
TAF5L functions as a fundamental unit of the transcription factor IID (TFIID) complex. This complex comprises several other TAF proteins and the TATA-binding protein (TBP) orchestrating the assembly of the preinitiation complex (PIC) on promoters. By stabilizing interactions between these proteins TAF5L facilitates the precise transcription initiation of various genes. Its role in the TFIID complex makes it essential for the recruitment of RNA polymerase II to specific DNA regions.
TAF5L actively participates in the RNA polymerase II transcription initiation pathway and the regulation of cellular response to DNA damage. Within these pathways its interaction with other TAF subunits and the TBP coordinates transcription activities. TAF5L specifically teams up with TAF6 to support the structural integrity of the TFIID complex ensuring effective transcription of genes in response to developmental and environmental signals.
TAF5L shows connections to certain cancers including colorectal and breast cancer. Alterations in TAF5L expression or function can lead to dysregulation of gene transcription contributing to tumorigenesis and cancer progression. The protein's association with the p53 signaling pathway responsible for maintaining genomic stability further highlights its importance in cancer. Additionally disruptions in TAF5L interactions with other TAF proteins or TBP may impact cell cycle control further linking it to cancer development.
