Function

The TFIID basal transcription factor complex plays a major role in the initiation of RNA polymerase II (Pol II)-dependent transcription (PubMed:33795473). TFIID recognizes and binds promoters with or without a TATA box via its subunit TBP, a TATA-box-binding protein, and promotes assembly of the pre-initiation complex (PIC) (PubMed:33795473). The TFIID complex consists of TBP and TBP-associated factors (TAFs), including TAF1, TAF2, TAF3, TAF4, TAF5, TAF6, TAF7, TAF8, TAF9, TAF10, TAF11, TAF12 and TAF13 (PubMed:33795473). TAF9 is also a component of the TBP-free TAFII complex (TFTC), the PCAF histone acetylase complex and the STAGA transcription coactivator-HAT complex (PubMed:15899866). TAF9 and its paralog TAF9B are involved in transcriptional activation as well as repression of distinct but overlapping sets of genes (PubMed:15899866). Essential for cell viability (PubMed:15899866). May have a role in gene regulation associated with apoptosis (PubMed:15899866).

Activity summary

TAF9 also known as TATA-box binding protein-associated factor 9 is a component of the transcription factor IID (TFIID) complex that plays an essential role in the initiation of gene transcription by RNA polymerase II. This protein has a molecular mass of around 28 kDa and is primarily located in the nucleus of the cell. TAF9 is expressed in various tissues with notable expression levels in liver and kidney.

Biological function summary

TAF9 associates with other proteins to form the TFIID complex an important component in the transcriptional machinery that binds to the TATA box region of the DNA. The interaction of TAF9 with TFIID facilitates the assembly of transcription initiation complexes at gene promoters impacting the transcription of numerous genes. This involvement in transcription regulation highlights TAF9's role in controlling cellular processes that depend on precise gene expression.

Pathways

TAF9 is actively involved in the RNA polymerase II transcription initiation pathway and the signaling pathway related to gene expression. Within these pathways TAF9 interacts with other TAF proteins such as TAF1 and TAF4 ensuring efficient transcription regulation. Its role connects with broader transcriptional networks that manage cellular function and response to environmental cues.

Associated diseases and disorders

Aberrant function or regulation of TAF9 is linked to liver cirrhosis and transcriptional dysregulation disorders. Changes in TAF9 expression can affect its interactions with components like TAF1 leading to disrupted transcriptional activities and potential disease progression. Understanding TAF9's connections to these diseases may offer insights for therapeutic strategies targeting transcription-related abnormalities.
