Activity summary

Talin 1 and Talin 2 also known as TLN1 and TLN2 are large cytoskeletal proteins with masses of approximately 270 kDa each. They act as mechanosensitive linkers that connect integrins to the actin cytoskeleton. Talins are critical for regulating cell adhesion migration and signaling. Both Talin 1 and 2 are expressed in various tissues including muscle heart and brain but their expression levels can differ depending on the tissue type.

Biological function summary

These proteins contribute to the formation of focal adhesions which are complexes that connect the extracellular matrix to actin. Talin engages with integrin receptors and activates them stabilizing integrin clustering and adhesion strength. Talin exists as part of a multiprotein complex that includes vinculin and paxillin which together control cellular responses to external mechanical forces.

Pathways

Talin 1 and 2 play central roles in integrin-mediated signaling pathways including the focal adhesion pathway and the PI3K/AKT pathway. Within these pathways Talins interact with integrin beta subunits and impact signaling cascades that manage cell survival proliferation and differentiation. Apart from integrins they associate with other proteins like kindlins which enhance integrin activation and clustering.

Associated diseases and disorders

Talin dysregulation associates with cancer metastasis and cardiovascular diseases. Aberrant Talin expression influences integrin signaling alterations linked to tumor progression and heart dysfunction. In cancer Talin modulates cellular invasion and migration interacting with proteins such as Src and FAK that are important for malignant cell behavior. In cardiovascular diseases altered Talin function can impair cardiac muscle integrity and contribute to pathological conditions.
