Supplementary information

TAM41 also known as TPRKB is a mitochondrial matrix protein with a molecular mass of approximately 41 kDa. It plays an important role in the biosynthesis of cardiolipin an important phospholipid for mitochondrial function. TAM41 is found mostly in the inner mitochondrial membrane where it actively participates in remodeling processes. Expression of TAM41 occurs in various tissues with the highest abundance in organs with high-energy demands like the heart and skeletal muscle.
Biological function summary

TAM41 is essential for proper mitochondrial maintenance by contributing to the cardiolipin biosynthesis pathway. This protein functions within a larger complex involved in lipid metabolism interacting with other proteins to facilitate the synthesis and integration of cardiolipin into the mitochondrial membrane. Through these actions TAM41 supports mitochondrial architecture and energy metabolism processes impacting cellular energy homeostasis.

Pathways

TAM41 operates within the cardiolipin biosynthesis and remodeling pathways. These pathways are integral to maintaining mitochondrial membrane stability and function. Interaction with proteins such as CRD1 and PGPS connects TAM41 to the essential mechanisms of lipid transport and assembly ensuring proper cellular energetics and mitochondrial dynamics.

Research links TAM41 to mitochondrial-related conditions particularly Barth syndrome. Mutations or dysregulation in TAM41 can compromise cardiolipin synthesis leading to cardiomyopathy and muscle weakness seen in these patients. Additionally interactions with CLD1 influence TAM41's involvement in conditions related to mitochondrial dysfunction highlighting its importance in maintaining mitochondrial integrity and health.
