Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) - Anti-TARC/CCL17 antibody [EPR15861] - BSA and Azide free (AB232659)

Immunohistochemical analysis of paraffin-embedded Human Hodgkin lymphoma tissue labeling TARC/CCL17 with ab195044 at 1/500 dilution, followed by Goat Anti-Rabbit IgG H&L (HRP) (ab97051) secondary antibody at 1/500 and counter-stained with Hematoxylin. Cytoplasm staining on humanHodgkin's lymphoma tissue was observed.
Inset : Negative control.

This data was developed using the same antibody clone in a different buffer formulation containing PBS, BSA, glycerol, and sodium azide (ab195044).

Heat mediated antigen retrieval was performed with Tris/EDTA buffer pH 9.0 before commencing with IHC staining protocol.

Product details

ab232659 is the carrier-free version of ab195044.

Patented technology
Our RabMAb® technology is a patented hybridoma-based technology for making rabbit monoclonal antibodies. For details on our patents, please refer to RabMAb® patents.

What are the advantages of a recombinant monoclonal antibody?
This product is a recombinant monoclonal antibody, which offers several advantages including:

  • - High batch-to-batch consistency and reproducibility
  • - Improved sensitivity and specificity
  • - Long-term security of supply
  • - Animal-free batch production

For more information, read more on recombinant antibodies.

Conjugation ready
Our carrier-free antibodies are typically supplied in a PBS-only formulation, purified and free of BSA, sodium azide and glycerol. This conjugation-ready format is designed for use with fluorochromes, metal isotopes, oligonucleotides, and enzymes, which makes them ideal for antibody labelling, functional and cell-based assays, flow-based assays (e.g. mass cytometry) and Multiplex Imaging applications.

Use our conjugation kits for antibody conjugates that are ready-to-use in as little as 20 minutes with 1 minute hands-on-time and 100% antibody recovery: available for fluorescent dyes, HRP, biotin and gold.

Compatibility
This product is compatible with the Maxpar® Antibody Labeling Kit from Fluidigm, without the need for antibody preparation. Maxpar® is a trademark of Fluidigm Canada Inc.

Supplementary information

TARC also known as CCL17 is a chemokine with a molecular mass of approximately 8 kDa. It is mainly expressed in the thymus but also found in the lung liver and lymph nodes. CCL17 plays an important role in immune response by signaling through the CCR4 receptor. It functions as a ligand that guides the movement of T-helper 2 (Th2) cells regulatory T cells (Tregs) and other immune cells to sites of inflammation.
Biological function summary

CCL17 acts as a chemotactic agent attracting specific immune cells especially T lymphocytes to inflammatory regions. This protein does not operate as part of a larger protein complex but engages directly by binding to receptors on the target immune cells. Localized expression of CCL17 increases in conditions where enhanced Th2 cell response is needed such as allergic or autoimmune responses.

Pathways

CCL17 plays a significant role in the cell signaling pathway called the chemokine signaling pathway. Through this pathway CCL17 interacts with the CCR4 receptor which helps mediate immune cell migration and adhesion. This interaction is critical for establishing the adaptive immune response. CCL17 and CCR4 support the trafficking of Th2 cells during immune reactions and pathogen defense.

CCL17 associates with asthma and atopic dermatitis where Th2 cell recruitment and activity are heightened. In asthma increased CCL17 levels exacerbate inflammatory response in the airways. In atopic dermatitis CCL17 presence corresponds with the intensity of the skin inflammation. These conditions are linked to the overactivity of Th2 cytokines such as IL-4 (H4L3) which collaborate with CCL17 to drive allergic responses.

Target data

Chemokine, which displays chemotactic activity for T lymphocytes, preferentially Th2 cells, but not monocytes or granulocytes. Therefore plays an important role in a wide range of inflammatory and immunological processes (PubMed : 8702936, PubMed : 9169480). Acts by binding to CCR4 at T-cell surface (PubMed : 10540332, PubMed : 9169480). Mediates GM-CSF/CSF2-driven pain and inflammation (PubMed : 27525438). In the brain, required to maintain the typical, highly branched morphology of hippocampal microglia under homeostatic conditions. May be important for the appropriate adaptation of microglial morphology and synaptic plasticity to acute lipopolysaccharide (LPS)-induced neuroinflammation (By similarity). Plays a role in wound healing, mainly by inducing fibroblast migration into the wound (By similarity).
