Supplementary information

TBC1D2 also known as Armus is a protein involved in tethering and trafficking of endosomes exhibiting a molecular mass of approximately 120 kDa. It interacts with Rab7 a small GTPase to regulate the elongation of endosomal structures. Expression of TBC1D2 is found in many tissues with significant levels in the brain and adipose tissue. The protein functions as a GAP (GTPase-activating protein) facilitating the conversion of Rab7-GTP to Rab7-GDP which is an important process in endosomal transport and maturation.
Biological function summary

TBC1D2 plays a role in the regulation of cellular autophagy and endocytosis. It is part of the endosomal complex involved in cargo sorting and trafficking affecting the lysosomal degradation pathway. The interaction with Rab7 suggests its involvement in the fusion of late endosomes with lysosomes. Additionally TBC1D2 regulates actin cytoskeleton dynamics by modulating signal transduction pathways impacting various cellular processes such as cell migration and morphogenesis.

Pathways

TBC1D2 participates in the endocytic and autophagic pathways influencing cellular homeostasis and response to stress. In the endocytic pathway it works closely with proteins like Rab7 to modulate endosome maturation. TBC1D2 also intersects with other autophagic-related proteins impacting cellular recycling processes important for maintaining cellular environment stability and responding to nutrient availability.

TBC1D2 is linked to neurodegenerative and metabolic conditions. Aberrant regulation of its activity is associated with Alzheimer's disease affecting amyloid precursor protein processing and clearance. Moreover TBC1D2's involvement in adipose tissue function connects it to obesity-related disorders influencing body weight and metabolism. Its interactions with proteins like Rab7 in these disease contexts underline its critical role in cellular trafficking and metabolic regulation.

Target data

Acts as a GTPase-activating protein for RAB7A. Signal effector acting as a linker between RAC1 and RAB7A, leading to RAB7A inactivation and subsequent inhibition of cadherin degradation and reduced cell-cell adhesion.
