TBC1D2B also known as TBC domain family member 2B is a Rab GTPase-activating protein (GAP) that plays an important role in regulating intracellular trafficking. The protein specifically accelerates the GTPase activity of Rab proteins which are key regulators of vesicle transport. TBC1D2B has a molecular mass of approximately 79 kDa and is widely expressed in various tissues including the brain liver and kidney.
Biological function summary

The TBC1D2B protein modulates vesicular traffic by acting as a GAP for Rab proteins specifically interacting with Rab7 a member of the Ras superfamily involved in endo-lysosomal trafficking. TBC1D2B acts as a negative regulator ensuring that the correct timing and specificity of vesicle fusion events are maintained. It functions as part of a complex with other proteins that together modulate endosome dynamics.

Pathways

TBC1D2B plays a significant role in the endocytosis pathway and the lysosome pathway.
