Supplementary information

The TBC1D5 protein also known as TBC1 domain family member 5 is involved in intracellular trafficking. It acts as a GTPase-activating protein (GAP) that regulates Rab GTPases. The molecular weight of TBC1D5 is approximately 76 kDa. This protein is widely expressed in various tissues with higher levels in neural tissues. Its presence suggests a role in cellular processes that require precise regulation of membrane trafficking and recycling dynamics.
Biological function summary

TBC1D5 regulates endosomal dynamics and is part of the retromer complex. It interacts with endosomal membranes helping to control the sorting and recycling of cargo proteins. The TBC1D5 protein modulates Rab7 and Rab9 activity by promoting GTP hydrolysis which influences vesicular trafficking. By coordinating with other retromer subunits like VPS29 and VPS35 TBC1D5 plays a significant role in maintaining cellular processes that require effective cargo transport and endosome maturation.

Pathways

TBC1D5 is closely linked to the endosomal trafficking and retrograde transport pathways. It connects functionally within the context of these pathways by interacting with Rab GTPases such as Rab7 and Rab9. TBC1D5 ensures proper endosome-to-Golgi transport modulating the dissociation and recycling of the retromer complex. Its actions influence how cargo is sorted and directed back to the trans-Golgi network impacting the downstream signaling events required for cellular homeostasis.

TBC1D5 has associations with neurodegenerative conditions like Parkinson's disease. In Parkinson's retromer dysfunction and disrupted endosomal sorting are observed implicating TBC1D5 in these pathological processes. Abnormalities in TBC1D5 function may lead to improper cargo sorting contributing to disease pathogenesis. The protein's relationship with VPS26 another retromer component plays a role in neurodegenerative disorder mechanisms highlighting a complex network affecting neuron survival and health.

Target data

May act as a GTPase-activating protein (GAP) for Rab family protein(s). May act as a GAP for RAB7A. Can displace RAB7A and retromer CSC subcomplex from the endosomal membrane to the cytosol; at least retromer displacement seems to require its catalytic activity (PubMed : 19531583, PubMed : 20923837). Required for retrograde transport of cargo proteins from endosomes to the trans-Golgi network (TGN); the function seems to require its catalytic activity. Involved in regulation of autophagy (PubMed : 22354992). May act as a molecular switch between endosomal and autophagosomal transport and is involved in reprogramming vesicle trafficking upon autophagy induction. Involved in the trafficking of ATG9A upon activation of autophagy. May regulate the recruitment of ATG9A-AP2-containing vesicles to autophagic membranes (PubMed : 24603492).
