Paraffin-embedded human cervical cancer tissue stained for TBCD/SSD1 using ab238761 at 1/300 dilution in immunohistochemical analysis.

After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0). Section was blocked with 10% normal goat serum 30 minutes at RT. Then primary antibody (1% BSA) was incubated at 4°C overnight. The primary is detected by a biotinylated secondary antibody and visualized using an HRP conjugated SP system.

TBCD also known as Tubulin Folding Cofactor D or SSD1 is a critical protein for tubulin folding and microtubule dynamics. It has a molecular mass of approximately 57 kDa and is located in various tissues with higher expression in the brain and testis. The protein works in combination with other tubulin cofactors to assist in proper β-tubulin folding and function which is important for maintaining cytoskeletal integrity.
Biological function summary

TBCD operates in conjunction with other proteins in a complex playing a significant role in microtubule biogenesis. It interacts with proteins such as TUBA1B to facilitate the proper folding and stability of β-tubulin. This process is essential for cell division and intracellular transport. Additionally it impacts neuronal structure by regulating axonal transport mechanisms influencing how neurons maintain their connections and communications.

Tubulin-folding protein implicated in the first step of the tubulin folding pathway and required for tubulin complex assembly. Involved in the regulation of microtubule polymerization or depolymerization, it modulates microtubule dynamics by capturing GTP-bound beta-tubulin (TUBB). Its ability to interact with beta tubulin is regulated via its interaction with ARL2. Acts as a GTPase-activating protein (GAP) for ARL2. Induces microtubule disruption in absence of ARL2. Increases degradation of beta tubulin, when overexpressed in polarized cells. Promotes epithelial cell detachment, a process antagonized by ARL2.
