TBLR1 also known as TBL1XR1 is a protein that plays an important role in transcription regulation mechanisms. This protein with a molecular mass of approximately 64 kDa is expressed in various tissues including the brain and lungs. It functions as an adaptor protein facilitating interactions between different transcription factors and the corepressor complex. By doing so it enables the repression or activation of specific genes according to cellular needs.
Biological function summary

TBLR1 participates in the regulation of gene expression by being part of the SMRT/N-CoR repressor complex. This complex interacts with nuclear receptors to modulate gene transcription. Through this function TBLR1 contributes to processes involved in maintaining cellular homeostasis. Its role in this complex marks it as a contributor to the fine-tuning of cellular responses to external stimuli which is essential for normal cellular function.

Pathways

The regulation of transcription by TBLR1 integrates into significant signaling pathways like Wnt and nuclear receptor pathways. It partners with proteins such as beta-catenin in the Wnt signaling pathway and corepressors in the nuclear receptor pathway. Through these partnerships TBLR1 affects important developmental and metabolic processes linking transcriptional machinery to broader regulatory networks in the cell.

TBLR1 has associations with conditions such as cancer and autism spectrum disorder. Mutations or dysregulation in the TBLR1 gene can affect normal cellular signaling leading to aberrant cell growth in cancers or disrupted neural development in autism. Additionally it is connected to the action of the protein N-CoR which is also implicated in these diseases. Through these associations TBLR1 becomes a target of interest for understanding and potentially treating these disorders.

F-box-like protein involved in the recruitment of the ubiquitin/19S proteasome complex to nuclear receptor-regulated transcription units. Plays an essential role in transcription activation mediated by nuclear receptors. Probably acts as integral component of the N-Cor corepressor complex that mediates the recruitment of the 19S proteasome complex, leading to the subsequent proteasomal degradation of N-Cor complex, thereby allowing cofactor exchange, and transcription activation.
