Biological function summary

TBX18 controls the regulation of cell differentiation and development especially in the heart and skeletal system. As a member of the T-box family of genes TBX18 participates in forming transcriptional regulation complexes. Its regulatory activities ensure that specific genes are expressed at the right time and location during developmental processes. This precise control is essential for normal embryonic development.

Activity summary

TBX18 also called T-box transcription factor Tbx18 is a transcription factor involved in DNA binding and gene regulation. It has a molecular mass of approximately 60 kDa. TBX18 expresses predominantly in developing tissues including the heart kidneys and limbs. It plays a role during embryogenesis and contributes to the regulation of developmental processes by modulating the expression of other genes important for organ formation.

Pathways

TBX18 integrates into the Wnt signaling and BMP pathways which are critical for cell fate determination and organogenesis. Within these pathways TBX18 interacts with other transcription factors like Tbx5 and Pitx2 to influence heart development. Its involvement in the pathways allows it to help steer mesodermal cells towards forming specific tissues showcasing its role in guiding developmental decisions.

Associated diseases and disorders

TBX18 has associations with congenital heart defects and limb malformations. Abnormal TBX18 expression or function can lead to structural heart anomalies with connections to other proteins like NKX2-5 and GATA4 which are also important in cardiac development.     ab115262 staining T-box transcription factor TBX18 in HeLa cells. The cells were fixed with 100% methanol (5 min), permeabilized with 0.1% PBS-Tween for 5 minutes and then blocked with 1% BSA/10% normal goat serum/0.3M glycine in 0.1%PBS-Tween for 1h. The cells were then incubated overnight at 4°C with ab115262 at 5 μg/ml and ab7291, Mouse monoclonal [DM1A] to alpha Tubulin - Loading Control. Cells were then incubated with ab150081, Goat polyclonal Secondary Antibody to Rabbit IgG - H&L (Alexa Fluor® 488), pre-adsorbed at 1/1000 dilution (shown in green) and ab150120, Goat polyclonal Secondary Antibody to Mouse IgG - H&L (Alexa Fluor® 594), pre-adsorbed at 1/1000 dilution (shown in pseudocolour red). Nuclear DNA was labelled with DAPI (shown in blue).

    Image was acquired with a high-content analyser (Operetta CLS, Perkin Elmer) and a maximum intensity projection of confocal sections is shown.

    This blot was produced using a 4-12% Bis-tris gel under the MOPS buffer system. The gel was run at 200V for 50 minutes before being transferred onto a Nitrocellulose membrane at 30V for 70 minutes. The membrane was then blocked for an hour using 5% Bovine Serum Albumin before being incubated with ab115262 overnight at 4°C. Antibody binding was detected using an anti-rabbit antibody conjugated to HRP, and visualised using ECL development solution.
