ab99302 staining Tbx3 in HepG2 cells. The cells were fixed with 4% Paraformaldehyde (10min), permeabilized with 0.1%PBS-Triton X-100 for 5 minutes and then blocked with 1% BSA/10% normal goat serum/0.3M glycine in 0.1%PBS-Tween for 1h. The cells were then incubated overnight at +4°C with ab99302 at 5μg/ml and ab7291, Mouse monoclonal [DM1A] to alpha Tubulin - Loading Control, at 1/1000 dilution. Cells were then incubated with ab150081, Goat polyclonal Secondary Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) pre-adsorbed, at 1/1000 dilution (shown in green) and ab150120, Goat polyclonal Secondary Antibody to Mouse IgG - H&L (Alexa Fluor® 594) pre-adsorbed, at 1/1000 dilution (shown in pseudocolor red). Nuclear DNA was labelled with DAPI (shown in blue).

Immunocytochemistry/ Immunofluorescence - Anti-Tbx3 antibody (AB99302)

ab99302 staining Tbx3 in the AR42J-B13 rat pancreatic tumor cell line by ICC/IF (Immunocytochemistry/immunofluorescence). Cells were fixed with paraformaldehyde, permeabilized with 0.1% Triton/PBS and blocked with blocking buffer for 60 minutes at room temperature. Samples were incubated with primary antibody (1/100) for 18 hours at 4°C. An Alexa Fluor® 488-conjugated goat anti-rabbit IgG polyclonal was used as the secondary antibody at a dilution of 1/5000.

Tbx3 also known as T-box transcription factor 3 is a transcription factor belonging to the T-box gene family. It has a molecular weight of approximately 74 kDa. The protein functions by binding to specific DNA sequences and regulating gene expression. It is involved in the repression or activation of target genes. Tbx3 is expressed in many tissues including the heart developing limbs and mammary glands.
Biological function summary

T-box transcription factor 3 is significant in embryonic development and cellular differentiation. It plays an important role in the development of the heart and limbs impacting cell fate decisions. Tbx3 interacts with other T-box family members and it is part of a regulatory network controlling pluripotency and differentiation in stem cells. Its expression patterns suggest important functions in organogenesis and tissue specification.

Tbx3 is a modulatory component in pathways controlling development and cell cycle progression. It is closely involved in the Wnt signaling and cell differentiation pathways. Transcriptional repressor involved in developmental processes (PubMed : 10468588). Binds to the palindromic T site 5'-TTCACACCTAGGTGTGAA-3' DNA sequence, or a half-site, which are present in the regulatory region of several genes (PubMed : 12000749). Probably plays a role in limb pattern formation (PubMed : 10468588). Required for mammary placode induction, and maintenance of the mammary buds during development (By similarity). Involved in branching morphogenesis in both developing lungs and adult mammary glands, via negative modulation of target genes; acting redundantly with TBX2 (By similarity). Required, together with TBX2, to maintain cell proliferation in the embryonic lung mesenchyme; perhaps acting downstream of SHH, BMP and TGFbeta signaling (By similarity). Involved in modulating early inner ear development, acting independently of, and also redundantly with, TBX2 in different subregions of the developing ear (By similarity).
