Probably plays a role in limb pattern formation (PubMed:10468588). Required for mammary placode induction, and maintenance of the mammary buds during development (By similarity). Involved in branching morphogenesis in both developing lungs and adult mammary glands, via negative modulation of target genes; acting redundantly with TBX2 (By similarity). Required, together with TBX2, to maintain cell proliferation in the embryonic lung mesenchyme; perhaps acting downstream of SHH, BMP and TGFbeta signaling (By similarity). Involved in modulating early inner ear development, acting independently of, and also redundantly with, TBX2 in different subregions of the developing ear (By similarity). Acts as a negative regulator of PML function in cellular senescence (PubMed:22002537).

Tbx3 also known as T-box transcription factor 3 is a transcription factor belonging to the T-box gene family. It has a molecular weight of approximately 74 kDa. The protein functions by binding to specific DNA sequences and regulating gene expression. It is involved in the repression or activation of target genes. Tbx3 is expressed in many tissues including the heart developing limbs and mammary glands.
T-box transcription factor 3 is significant in embryonic development and cellular differentiation. It plays an important role in the development of the heart and limbs impacting cell fate decisions. Tbx3 interacts with other T-box family members and it is part of a regulatory network controlling pluripotency and differentiation in stem cells. Its expression patterns suggest important functions in organogenesis and tissue specification.
Tbx3 is a modulatory component in pathways controlling development and cell cycle progression. It is closely involved in the Wnt signaling and cell differentiation pathways. In the Wnt pathway Tbx3 regulates genes essential for cell proliferation and development. It interacts with proteins such as β-catenin linking it to other pathway members like LEF/TCF transcription factors further influencing cellular responses and development processes.
T-box transcription factor 3 has associations with certain cancers and developmental disorders. Mutations in Tbx3 can lead to ulnar-mammary syndrome a disorder that affects limb and mammary gland development. In cancer Tbx3 overexpression is linked to melanoma progression interacting with proteins like p53 to affect cell cycle regulation and apoptotic pathways. Understanding Tbx3's role may provide insights into therapeutic targets for these conditions.
