TC1 also known as Transcriptional Coactivator 1 is a protein with a molecular mass approximately 55 kDa. It plays a role in gene expression. TC1 commonly influences transcription by interacting with transcription factors and the basal transcription machinery. The protein is mostly expressed in the nucleus of cells especially in tissues with high rates of cell proliferation like the liver and spleen. TC1 affects transcription of several genes by modifying chromatin structure and recruiting other proteins necessary for gene expression.
Biological function summary

TC1 facilitates the transcriptional activity of various genes essential for cell cycle regulation and differentiation. The protein participates in multi-protein complexes forming part of a larger assembly to enhance gene expression efficiency. Through its involvement in the regulation of cell cycle-related genes TC1 impacts cellular processes like growth proliferation and apoptosis. It modulates gene expression by restructuring chromatin to allow transcription factors and related proteins better access to DNA sequences.

Pathways

TC1 integrates into the cell cycle control and apoptosis regulation networks. It interacts with proteins such as Cyclin D playing a role in the G1/S phase transition and with members of the E2F transcription factor family which are key for DNA synthesis control. TC1's activities significantly impact pathways like the retinoblastoma (Rb) pathway which governs cell cycle progression and it aligns with proteins that regulate cell death and survival balances ensuring proper cell function and organismal homeostasis.

TC1 shows strong connections to cancer and liver disorders. Overexpression or malfunction of TC1 has been linked to increased cell proliferation and tumorigenesis especially in cancers like hepatocellular carcinoma. Aberrant TC1 activity influences Cyclin D and other cell cycle proteins contributing to uncontrolled cell division and tumor progression. Additionally irregular TC1 expression interacts with the Rb protein pathway further implicating it in the development and maintenance of these disorders.
