The TCF-4/TCF7L2 and TCF-3/TCF7L1 proteins are part of the T-cell factor/lymphoid enhancer-binding factor (TCF/LEF) family. These proteins also known as transcription factor 7 like 2 and transcription factor 7 like 1 function as transcription factors in the Wnt signaling pathway. TCF7L2 and TCF7L1 typically weigh around 65 kDa and 56 kDa respectively. Expression of these proteins occurs in many tissues notably in the colon liver and pancreas. Mechanically they interact with β-catenin to regulate the transcription of Wnt target genes.
Biological function summary

TCF-4/TCF7L2 and TCF-3/TCF7L1 play key roles in controlling gene expression essential for cell proliferation differentiation and embryonic development. These transcription factors often form complexes with β-catenin coactivators or corepressors to modulate gene transcription activity. By influencing genetic programs they contribute to developmental processes and the maintenance of stem cells' properties. Their involvement in embryogenesis highlights their importance in developmental signaling.

Pathways

TCF-4/TCF7L2 and TCF-3/TCF7L1 are significant in the canonical Wnt/β-catenin signaling pathway. This pathway influences many cellular processes including cell fate and cellular proliferation decisions. When Wnt ligands activate the pathway β-catenin accumulates in the cytoplasm and enters the nucleus where it partners with TCF proteins to modify gene expression. This interaction highlights the relationship of TCF proteins with β-catenin and other Wnt-associated elements like Axin and APC which regulate β-catenin stability.

Alterations in TCF7L2 and TCF7L1 activity impact conditions like colorectal cancer and type 2 diabetes. Mutations or misexpression of TCF7L2 show strong links to increased risk of type 2 diabetes across various populations. In colorectal cancer changes in Wnt signaling and TCF's role as transcription factors contribute to oncogenesis. TCF proteins interact with other factors in these contexts such as the adenomatous polyposis coli (APC) protein highlighting their relevance in disease pathology.

Target data

Participates in the Wnt signaling pathway and modulates MYC expression by binding to its promoter in a sequence-specific manner. Acts as a repressor in the absence of CTNNB1, and as activator in its presence. Activates transcription from promoters with several copies of the Tcf motif CCTTTGATC in the presence of CTNNB1. TLE1, TLE2, TLE3 and TLE4 repress transactivation mediated by TCF7L2/TCF4 and CTNNB1. Expression of dominant-negative mutants results in cell-cycle arrest in G1 (By similarity). Necessary for the maintenance of the epithelial stem-cell compartment of the small intestine.
