TCP1 epsilon also known as CCT5 is an important component of the chaperonin-containing TCP1 complex. This protein weighs around 61 kDa and is expressed in a variety of tissues with higher prevalence in areas with significant protein synthesis and folding activities like the brain and testis. Mechanically TCP1 epsilon facilitates the folding of newly synthesized and stress-denatured proteins by providing an optimal environment for proper protein conformation. It does this by assembling into a barrel-shaped structure that encapsulates the substrate proteins.
Biological function summary

CCT5 is part of the cytosolic chaperonin complex called the TRiC (TCP1 Ring Complex) which is essential for the folding of actin and tubulin. This complex ensures that cytoskeletal proteins achieve their correct tertiary structures necessary for maintaining cellular architecture and function. Additionally this complex prevents the misfolding and aggregation of proteins a process that helps maintain cellular homeostasis and prevent cellular stress.

Pathways

CCT5 plays a significant role in the cytoskeleton modeling and protein homeostasis pathways. In the cytoskeleton pathway CCT5's interaction with actin and tubulin is vital ensuring these proteins attain functional conformation. In the protein homeostasis pathway CCT5 assists in degrading misfolded proteins working closely with other components like heat shock proteins (HSPs) to manage cellular protein load effectively. This interaction highlights its importance in preventing the accumulation of damaged proteins.

Mutations in CCT5 have been linked to some neurodegenerative conditions like Hereditary Sensory Neuropathy Type 2 (HSN2). The failure of CCT5 to assist in proper protein folding can lead to neuronal damage owing to protein aggregates. Additionally disruptions in CCT5 are associated with some forms of cancer where abnormal protein folding influences cell proliferation. CCT5 work in conjunction with other proteins like p53 to maintain cellular function and dysregulation in cancer highlights its role in cellular growth and apoptosis regulation.
