TRBC1 also known as T-Cell Receptor Beta Constant 1 is involved in the formation of the T-cell receptor complex. It has a molecular mass of around 18 kDa. This protein component is prominently expressed on the surface of T lymphocytes important for mediating immune responses. TRBC1 is a part of the TCR constant region playing a significant role in antigen recognition through its interaction with antigen-presenting cells.
Biological function summary

TRBC1 contributes to the immune system’s mechanism by helping identify foreign invaders in the body. It works as an integral part of the T-cell receptor (TCR) complex facilitating T-cell activation and signaling. The complex formation includes variable and constant regions with TRBC1 as a fixed part of the constant region ensuring the proper receptor structure for effective recognition. TCR assembly is important for T cells to detect specific antigens.

Pathways

TRBC1 participates in the adaptive immune response and T-cell receptor signaling pathway. Interacting proteins such as CD3 and ZAP70 help transmit signals initiated at the surface by antigen recognition leading to downstream effects such as cell proliferation and cytokine release. TRBC1’s involvement in these pathways ensures the T cells' ability to respond to invading pathogens and maintain immune surveillance.

TRBC1 has been implicated in conditions like autoimmune diseases and certain cancers such as T-cell lymphomas. Aberrant TRBC1 expression or mutations can disrupt normal immune functions triggering disease development or progression. Target data

Constant region of T cell receptor (TR) beta chain (PubMed : 24600447). Alpha-beta T cell receptors are antigen specific receptors which are essential to the immune response and are present on the cell surface of T lymphocytes. Recognize peptide-major histocompatibility (MH) (pMH) complexes that are displayed by antigen presenting cells (APC), a prerequisite for efficient T cell adaptive immunity against pathogens (PubMed : 25493333). Binding of alpha-beta TR to pMH complex initiates TR-CD3 clustering on the cell surface and intracellular activation of LCK that phosphorylates the ITAM motifs of CD3G, CD3D, CD3E and CD247 enabling the recruitment of ZAP70. In turn, ZAP70 phosphorylates LAT, which recruits numerous signaling molecules to form the LAT signalosome. The LAT signalosome propagates signal branching to three major signaling pathways, the calcium, the mitogen-activated protein kinase (MAPK) kinase and the nuclear factor NF-kappa-B (NF-kB) pathways, leading to the mobilization of transcription factors that are critical for gene expression and essential for T cell growth and differentiation (PubMed : 23524462, PubMed : 9382891). The T cell repertoire is generated in the thymus, by V-(D)-J rearrangement. This repertoire is then shaped by intrathymic selection events to generate a peripheral T cell pool of self-MH restricted, non-autoaggressive T cells. Post-thymic interaction of alpha-beta TR with the pMH complexes shapes TR structural and functional avidity (PubMed : 15040585).
