Tectonic family member 1 (TCTN1) also known as protein tectonic-1 is an important component of cellular function. Its molecular weight is approximately 63 kilodaltons. Primarily TCTN1 is expressed in ciliated cells and other tissues where primary cilia functions are maintained. The protein localizes to the ciliary transition zone playing an important role in ciliary development and function which makes it an important structural constituent regulating the ciliary membrane composition.
Biological function summary

TCTN1 associates with multiple ciliary proteins forming part of the tectonic complex. This complex stabilizes the transition zone in primary cilia acting as a barrier that controls the protein and ion passage between the cilia and cytoplasm. TCTN1’s interaction with other members of the tectonic family helps maintain ciliary structure and functionality which are critical in signal transduction pathways implicated in development and cell cycle regulation.

Pathways

TCTN1 functions integrally in the Hedgehog signaling pathway and is involved in Wnt signaling to some extent. This positions TCTN1 as a modulator in these pathways influencing cellular responses important for growth and differentiation. Its interaction with other proteins like GLI family members and SMO (Smoothened) highlights its role in Hedgehog signaling modulation particularly affecting gene transcription processes that direct embryonic patterning and tissue homeostasis.

TCTN1 has connections to Joubert Syndrome and Meckel-Gruber Syndrome both ciliopathies marked by dysfunctional cilia. Mutations or functional impairments in TCTN1 compromise cilia structure and signaling processes contributing to the pathogenesis of these conditions. The protein’s interaction with MKS1 and CEP290 is critical as disruptions here are noted in aforementioned disorders highlighting its importance in maintaining ciliary integrity for preventing disease.

Component of the tectonic-like complex, a complex localized at the transition zone of primary cilia and acting as a barrier that prevents diffusion of transmembrane proteins between the cilia and plasma membranes. Regulator of Hedgehog (Hh), required for both activation and inhibition of the Hh pathway in the patterning of the neural tube. During neural tube development, it is required for formation of the most ventral cell types and for full Hh pathway activation. Functions in Hh signal transduction to fully activate the pathway in the presence of high Hh levels and to repress the pathway in the absence of Hh signals. Modulates Hh signal transduction downstream of SMO and RAB23 (By similarity).
