Flow cytometric analysis of 4% paraformaldehyde fixed 90% methanol permeabilized Jurkat cells labelling TCTP with ab124835 at 1/20 dilution (7.5 µg) (Red) compared with a Rabbit monoclonal IgG (Black) isotype control and an unlabelled control (cells without incubation with primary antibody and secondary antibody) (Blue). Goat Anti-Rabbit IgG (Alexa Fluor® 488 ab150081) at 1/5000 dilution was used as the secondary antibody.

Immunofluorescent analysis of 4% Paraformaldehyde-fixed 0.1% TritonX-100 permeabilized Jurkat cells with ab124835 at 1/50 (3.08 µg/ml) dilution followed by ab150081 Goat Anti-Rabbit IgG H&L (Alexa Fluor® 488) preadsorbed antibody at 1/1000 (2 µg/ml) dilution (Green). Confocal image showing cytoplasmic staining in Jurkat cells. Anti-alpha Tubulin mouse monoclonal antibody - Microtubule Marker (Alexa Fluor® 594) was used to counterstain tubulin at 1/200 (2.5ug/ml) dilution (Red). The Nuclear counterstain was DAPI (Blue).

TCTP also known as Translationally Controlled Tumor Protein or histamine-releasing factor (HRF) is a multifunctional protein with a molecular mass of approximately 19 kDa. It is found in a variety of tissues and cell types including cytoplasm and extracellular environments. The protein engages in cellular processes such as cell growth development and apoptosis. TCTP regulates microtubule stability cell cycle progression and protein synthesis reflecting its broad mechanical roles within cells.
Biological function summary

In both animal and human cells TCTP acts as a prosurvival factor by inhibiting apoptosis. It binds with proteins like Bcl-2 and Bax playing a role in the regulation of cell death. Furthermore TCTP appears to mediate immune responses through its histamine-releasing factor activity. It interacts within protein complexes that manage cytoskeletal dynamics and stress responses contributing to cellular adaptability.

Pathways

The protein interacts in critical pathways such as the MAPK signaling pathway and the apoptosis pathway. In the MAPK pathway TCTP modulates cell growth and proliferation by interacting with proteins like ERK1/2.
