Tethering factor involved in autophagy. Involved in autophagosome maturation by promoting the autophagosome fusion with lysosomes: acts by associating with both the ATG5-ATG12 conjugate and phosphatidylinositol-3-phosphate (PtdIns(3)P) present at the surface of autophagosomes. TECPR1 also known as Tectonin Beta-Propeller Repeat Containing 1 is a protein playing an important role in autophagy. It has a molecular mass of approximately 96 kDa. TECPR1 is widely expressed in human tissues including the brain liver and skeletal muscles. This protein contains multiple tectonin beta-propeller repeats believed to help in forming protein-protein interactions essential for its function.
The TECPR1 protein assists in the regulation and execution of the autophagy process particularly in selective autophagy or aggrephagy. By functioning as part of a protein complex it connects with components of the autophagy machinery supporting the clearance of damaged organelles and aggregation-prone proteins. Its role in maintaining cellular homeostasis is critical and indicates its involvement in the cellular defense against stress conditions.
TECPR1 engages notably in the autophagy pathway interacting with important proteins like LC3 and ATG5. This protein is also associated with the formation of autophagosomes and their fusion with lysosomes during the degradation process. TECPR1 contributes to the maintenance of protein quality control sharing connections with other autophagic proteins necessary for these pathways.
TECPR1 mutations have connections to neurodegenerative conditions such as hereditary spastic paraplegia. These genetic changes can disrupt the autophagic process leading to the accumulation of dysfunctional proteins and organelles therefore contributing to neuronal damage. TECPR1 also relates to lysosomal storage disorders where its interaction with proteins like p62/SQSTM1 is notable showing its role in pathogenic processes involving cellular cleansing failure and aggregate formation.
