Immunocytochemistry/ Immunofluorescence - Anti-TEM8/ATR antibody [EPNCI-R173-37] - BSA and Azide free (AB246321)

Immunofluorescent analysis of 100% Methanol fixed LoVo (human colorectal adenocarcinoma epithelial cell) cells labeling TEM8/ATR with ab241067 at 1/100 dilution (Green), followed by Goat Anti-Rabbit IgG H&L (Alexa Fluor® 488) (ab150077) secondary antibody at 1/1000 dilution. Confocal image showing cytoplasmic and plasma membrane staining in LoVo cells.

The nuclear counter stain is DAPI (blue). Tubulin is detected with Anti-alpha Tubulin antibody [DM1A] - Microtubule Marker (Alexa Fluor® 594) (ab195889) (red) at 1/200 dilution.

Secondary antibody only control : Used PBS instead of primary antibody, secondary antibody is Goat Anti-Rabbit IgG H&L (Alexa Fluor® 488) (ab150077) secondary antibody at 1/1000 dilution.

PFA fixation is not recommended due to weak staining on LoVo cells.

This data was developed using the same antibody clone in a different buffer formulation containing PBS, BSA, glycerol, and sodium azide (ab241067).

Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) - Anti-TEM8/ATR antibody [EPNCI-R173-37] - BSA and Azide free (AB246321)

Immunohistochemical analysis of 10% neutral-buffered formalin Mammary fat pad of TEM8/ATR wildtype (WT) or knockout (KO) athymic nude mice transplanted with HT29 human colon tumor tissue labeling TEM8/ATR with ab241067 at 1/60 dilution, followed by a Biotin anti-rabbit IgG secondsry antibody. Positive staining in stromal cells of transplanted HT29 human colon tumor growing in TEM8/ATR wildtype (WT) athymic nude mice is observed. Antigen retrieval : Retrieved with proteinase K. This image was kindly provided by our collaborator Dr. Brad St. Croix, National Cancer Institute (NCI), NIH, and published in J Clin Invest. 128 : 2927-2943 (PMID : 29863500). This data was developed using the same antibody clone in a different buffer formulation containing PBS, BSA, glycerol, and sodium azide (ab241067).

Product details

ab246321 is the carrier-free version of ab241067.

Conjugation ready
Our carrier-free antibodies are typically supplied in a PBS-only formulation, purified and free of BSA, sodium azide and glycerol. This conjugation-ready format is designed for use with fluorochromes, metal isotopes, oligonucleotides, and enzymes, which makes them ideal for antibody labelling, functional and cell-based assays, flow-based assays (e.g. mass cytometry) and Multiplex Imaging applications.

Use our conjugation kits for antibody conjugates that are ready-to-use in as little as 20 minutes with 1 minute hands-on-time and 100% antibody recovery: available for fluorescent dyes, HRP, biotin and gold.

Compatibility
This product is compatible with the Maxpar® Antibody Labeling Kit from Fluidigm, without the need for antibody preparation. Maxpar® is a trademark of Fluidigm Canada Inc.

Target data

Plays a role in cell attachment and migration. Interacts with extracellular matrix proteins and with the actin cytoskeleton. Mediates adhesion of cells to type 1 collagen and gelatin, reorganization of the actin cytoskeleton and promotes cell spreading. Plays a role in the angiogenic response of cultured umbilical vein endothelial cells.. (Microbial infection) Acts as a receptor for protective antigen (PA) of B.anthracis.
