Supplementary information

Tenascin-N also known as TNN or Tenascin-W is a glycoprotein within the tenascin family which includes Tenascin-C Tenascin-X and Tenascin-R. It weighs approximately 150 kDa. Tenascin-N is expressed in specific tissues including the central nervous system muscle tissues and various regions of the developing skeleton. It mainly localizes in the extracellular matrix where it interacts with components like fibronectin and integrins to facilitate cellular interactions.
Biological function summary

The protein plays important roles in cell adhesion migration and proliferation. It does not typically form part of a larger complex but acts independently to modulate cellular environments. In the context of musculoskeletal development and neural tissue architecture Tenascin-N creates a scaffold that supports cell organization and tissue rigidity. It adjusts cell behavior through binding interactions impacting essential developmental signaling.

Pathways

Tenascin-N influences specific signaling cascades pertinent to skeletal and nervous system development. It interacts with the Wnt signaling pathway aligning with proteins such as β-catenin. Furthermore it modulates the PI3K/AKT pathway influencing cell survival and proliferation. These interactions reflect Tenascin-N's role in development and response to cellular stress.

Tenascin-N associates with gliomas and osteosarcomas. In glial tumors its expression levels correlate with tumor progression possibly interacting with integrin-linked proteins. Osteosarcomas display altered Tenascin-N patterns indicating a connection to abnormal osteoblast function. In these contexts Tenascin-N works alongside other tenascin family members fostering environments conducive to malignant growth and altered cellular mechanics.

Target data

Extracellular matrix protein that seems to be a ligand for ITGA8 : ITGB1, ITGAV : ITGB1 and ITGA4 : ITGB1 (By similarity) (PubMed : 17909022). Involved in neurite outgrowth and cell migration in hippocampal explants (By similarity). During endochondral bone formation, inhibits proliferation and differentiation of proteoblasts mediated by canonical WNT signaling (By similarity). In tumors, stimulates angiogenesis by elongation, migration and sprouting of endothelial cells (PubMed : 19884327). Expressed in most mammary tumors, may facilitate tumorigenesis by supporting the migratory behavior of breast cancer cells (PubMed : 17909022).
