TetR or tetracycline repressor protein is a regulatory protein that controls the expression of genes involved in resistance to tetracycline. It weighs about 23 kDa. TetR functions as a homodimer binding to substrate DNA sequences to suppress transcription. This repression occurs by binding to the operator sequences of the tetracycline-inducible promoter blocking the progression of RNA polymerase along the DNA strand. TetR mainly operates within the bacterial species Escherichia coli serving as a well-characterized model for gene regulation in prokaryotes.
Biological function summary

TetR plays a critical role in antibiotic resistance mechanisms. When tetracycline is not present TetR maintains its binding to the operator effectively silencing the expression of downstream genes responsible for drug efflux. Upon binding tetracycline the protein undergoes a conformational change that releases the operator sequence allowing the transcription of resistance genes.
