TetR or tetracycline repressor protein is a regulatory protein that controls the expression of genes involved in resistance to tetracycline. It weighs about 23 kDa. TetR functions as a homodimer binding to substrate DNA sequences to suppress transcription. This repression occurs by binding to the operator sequences of the tetracycline-inducible promoter blocking the progression of RNA polymerase along the DNA strand. TetR mainly operates within the bacterial species Escherichia coli serving as a well-characterized model for gene regulation in prokaryotes.
TetR plays a critical role in antibiotic resistance mechanisms. When tetracycline is not present TetR maintains its binding to the operator effectively silencing the expression of downstream genes responsible for drug efflux. Upon binding tetracycline the protein undergoes a conformational change that releases the operator sequence allowing the transcription of resistance genes. TetR forms a complex through dimerization which is essential for its function as it binds the operator in pairs.
TetR is integral to the regulation of the tetracycline resistance pathway. It is part of a classic feedback loop that includes TetA a protein involved in tetracycline efflux out of the cell. The TetR-TetA regulatory system finely tunes bacterial response to the presence or absence of tetracycline ensuring survival in antibiotic environments. Its interaction with the TetA protein highlights its essential role in regulating efflux pump expression during antibiotic exposure.
TetR connects primarily to antibiotic resistance a major issue in bacterial infections. The misuse of tetracycline in clinical settings emphasizes the significance of TetR in developing tetracycline-resistant bacterial strains. This protein's connection to the TetA efflux pump directly affects the efficacy of tetracycline as an antibacterial treatment. Understanding the TetR regulation mechanism could lead to new therapeutic strategies in combating drug-resistant pathogens particularly methicillin-resistant Staphylococcus aureus (MRSA).

Immunofluorescent analysis of 4% Paraformaldehyde-fixed, 0.1% TritonX-100 permeabilized HEK293T (human embryonic kidney epithelial cell) cells labelling TetR with Anti-TetR antibody [31B3] ab302642 at 1/50 (17.58 ug/mL) dilution, followed by Goat Anti-Mouse IgG H&L (Alexa Fluor® 488) preadsorbed ab150117 Goat Anti-Mouse IgG H&L (Alexa Fluor® 488) preadsorbed antibody at 1/1000 2ug/mL dilution (Green).
