Biological function summary
The role of TH1L extends beyond just a mechanical scaffold. As part of the mediator complex TH1L plays an important role in regulating gene expression by interacting with transcriptional activators and repressors. It integrates signals from transcription factors and conveys them to the transcriptional machinery thereby influencing which genes are turned on or off. TH1L is instrumental in the coordinated control of gene expression impacting diverse biological processes such as cell growth differentiation and response to external stimuli.
Pathways
TH1L's involvement is significant in the Notch signaling pathway and the Wnt signaling pathway both important for cell fate determination and proliferation. In the Notch pathway TH1L helps regulate the transcription of target genes by modulating the activity of the Notch intracellular domain. In the Wnt signaling TH1L assists in the transcriptional regulation by interacting with transcription factors like TCF/LEF that respond to Wnt signals. These pathways form a complex network of protein interactions with TH1L working in conjunction with other proteins like SMAD and β-catenin to fine-tune gene expression outcomes.
