Product details

ab249073 is the carrier-free version of ab154173.

Patented technology
Our RabMAb® technology is a patented hybridoma-based technology for making rabbit monoclonal antibodies. For details on our patents, please refer to RabMAb® patents.

What are the advantages of a recombinant monoclonal antibody?
This product is a recombinant monoclonal antibody, which offers several advantages including:

  • - High batch-to-batch consistency and reproducibility
  • - Improved sensitivity and specificity
  • - Long-term security of supply
  • - Animal-free batch production

For more information, read more on recombinant antibodies.

Conjugation ready
Our carrier-free antibodies are typically supplied in a PBS-only formulation, purified and free of BSA, sodium azide and glycerol. This conjugation-ready format is designed for use with fluorochromes, metal isotopes, oligonucleotides, and enzymes, which makes them ideal for antibody labelling, functional and cell-based assays, flow-based assays (e.g. mass cytometry) and Multiplex Imaging applications.

Use our conjugation kits for antibody conjugates that are ready-to-use in as little as 20 minutes with 1 minute hands-on-time and 100% antibody recovery: available for fluorescent dyes, HRP, biotin and gold.

Compatibility
This product is compatible with the Maxpar® Antibody Labeling Kit from Fluidigm, without the need for antibody preparation. Maxpar® is a trademark of Fluidigm Canada Inc.

Supplementary information

Thimet Oligopeptidase also known as EC 3.4.24.15 or TOP functions as a zinc-dependent metallopeptidase. It breaks down peptides that are up to 20 amino acids long by cleaving peptide bonds. It has a molecular mass of approximately 78 kDa. This enzyme is expressed in various tissues prominently in the brain but also found in the kidneys lungs and testis reflecting its broad physiological roles. TOP primarily exists in the cytosol but can translocate to other cellular compartments under specific conditions.
Biological function summary

Thimet Oligopeptidase plays a significant role in modulating neuropeptide levels in the nervous system. It does not operate as part of a known complex but instead acts independently in intracellular peptide catabolism. This activity helps regulate processes such as pain modulation and neuroendocrine signaling by modifying peptide-based signaling molecules. Furthermore TOP participates in the degradation of antigenic peptides implicating a role in immune system regulation.

Pathways

Thimet Oligopeptidase participates in neuropeptide and peptide hormone metabolism where it helps maintain homeostasis by modulating bioactive peptide concentrations. Within these pathways TOP interacts with other proteins such as neprilysin and angiotensin-converting enzyme creating a network that helps regulate cardiovascular and neural functions. This interaction highlights the enzyme's versatile capability to influence multiple signaling cascades.

Thimet Oligopeptidase connects strongly to neurodegenerative diseases such as Alzheimer's and Parkinson's. Research indicates that abnormal activity of TOP contributes to the accumulation of toxic peptide fragments like amyloid-beta in Alzheimer’s disease. This connection brings it into association with proteins like amyloid precursor protein (APP) and tau which are key in such neurodegenerative conditions. The enzyme's regulation of peptidase activity in these contexts highlights its potential as a therapeutic target for disease intervention.

Target data

Involved in the metabolism of neuropeptides under 20 amino acid residues long. Involved in cytoplasmic peptide degradation (PubMed : 17251185, PubMed : 7639763). Able to degrade the amyloid-beta precursor protein and generate amyloidogenic fragments (PubMed : 17251185, PubMed : 7639763). Also acts as a regulator of cannabinoid signaling pathway by mediating degradation of hemopressin, an antagonist peptide of the cannabinoid receptor CNR1 (By similarity).
