THOC2 also known as Tho2 is part of the THO complex and is an important player in mRNA processing and transport. This protein has an estimated mass of around 200 kDa and is mainly expressed in the nucleus of eukaryotic cells. THOC2 plays a significant role in RNA metabolism binding to nascent mRNA and helping with the integration of several other factors needed for RNA stability and transport to the cytoplasm.
Biological function summary

THOC2 is necessary for efficient mRNA export and is part of the larger transcription export (TREX) complex which involves several proteins including THOC1 THOC5 and UAP56. This complex couples transcription with mRNA processing contributing to the maturation and export of mRNA transcripts. THOC2 ensures the proper processing and exportation of mRNA preventing the accumulation of ribonucleoprotein complexes in the nucleus.

Pathways

THOC2 integrates into the process of gene expression where it links transcription with mRNA export. It participates in pathways related to RNA binding and export. During this THOC2 interacts with proteins such as ALYREF and SR proteins which further support the processes by facilitating efficient mRNA transport and splicing. Additionally the presence of THOC2 in the THO complex is essential for the stability of the complex influencing the overall gene expression regulation.

THOC2 mutations have associations with intellectual disabilities and other neurological issues. Mutations in the THOC2 gene can disrupt its normal function in RNA processing leading to deficits in brain development and function. Furthermore THOC2's connection with other THO complex proteins involves interactions implicated in these disorders. For example disruptions that affect the stability or function of other TREX components like UAP56 can amplify disease phenotypes by disturbing mRNA transport and processing.

Component of the THO subcomplex of the TREX complex which is thought to couple mRNA transcription, processing and nuclear export, and which specifically associates with spliced mRNA and not with unspliced pre-mRNA (PubMed : 15833825, PubMed : 15998806, PubMed : 17190602). Required for efficient export of polyadenylated RNA and spliced mRNA (PubMed : 23222130). The THOC1-THOC2-THOC3 core complex alone is sufficient to bind export factor NXF1-NXT1 and promote ATPase activity of DDX39B; in the complex THOC2 is the only component that directly interacts with DDX39B (PubMed : 33191911). TREX is recruited to spliced mRNAs by a transcription-independent mechanism, binds to mRNA upstream of the exon-junction complex (EJC) and is recruited in a splicing- and cap-dependent manner to a region near the 5' end of the mRNA where it functions in mRNA export to the cytoplasm via the TAP/NXF1 pathway (PubMed : 15833825, PubMed : 15998806, PubMed : 17190602). Required for NXF1 localization to the nuclear rim (PubMed : 22893130). THOC2 (and probably the THO complex) is involved in releasing mRNA from nuclear speckle domains..
