Supplementary information

The Thomsen-Friedenreich antigen (TF antigen) also known as CD176 is a cryptic carbohydrate antigen exposed on the surface of cells particularly after malignant transformation. It has a molecular mass that varies depending on the glycoprotein carrier. TF antigen often exists in an inactive form within normal epithelial and endothelial cells masked by sialic acid and becomes exposed in cancerous and certain pathological conditions. Its structural core consists of a galactose beta 1-3 N-acetylgalactosamine configuration.
Biological function summary

The TF antigen plays roles in cell adhesion signal transduction and as a marker of cancer progression. It is not part of a larger protein complex but serves a significant role by participating in cellular recognition and immune response evasion. Researchers identified it as a pan-carcinoma antigen often used as a target for cancer immunotherapy due to its expression in many tumor tissues such as breast and colorectal cancers.

Pathways

TF antigen is involved in processes related to cell adhesion and immune evasion. These interactions often include pathways like the PI3K/AKT signaling pathway which contribute to cell survival and proliferation and involve proteins such as integrins and lectins. The alteration in pathways due to TF expression changes helps cancer cells migrate and grow making it an important target for analyzing tumor behavior.

TF antigen closely associates with cancer specifically aiding in tumorigenesis and metastasis in breast and colorectal cancers. Its expression correlates with proteins like MUC1 and MUC2 which are mucins often overexpressed and modified in these cancers. The exposure of TF antigen on these mucins can be a marker for malignancy and the interaction further disrupts normal cell function contributing to disease progression.
