Immunofluorescence analysis of paraformaldehyde fixed mouse splenocytes on Shi-fix® coverslips stained with the chimeric ab281910 at 10 μg/ml for 1 hour, followed by Alexa Fluor® 488 secondary antibody at 2 μg/ml (green/top-left). Image showing membrane staining. The nuclear counter stain is DAPI (blue). The isotype control was an unknown specificity antibody followed by staining with Alexa Fluor® 488 secondary antibody. Mouse splenocytes were immobilized on Shi-fix® coverslips.

Biological function summary
Tim-2 plays an important role in immune regulation and functions as a part of the broader family of T-cell immunoglobulin and mucin domain proteins. It does not typically form part of a complex but interacts with phosphatidylserine a phospholipid exposed during cell apoptosis. This interaction suggests Tim-2 has roles in recognizing apoptotic cells and may influence their removal by macrophages. Additionally it affects lymphocyte activity possibly by modulating T-cell proliferation and cytokine release.
Pathways
Tim-2 influences immune responses and cell survival pathways. It connects to the phosphatidylserine-mediated clearance pathway a critical component of maintaining tissue homeostasis and preventing inflammation by promoting the clearance of dying cells. Tim-2 also interacts with related proteins such as Tim-3 which shares similar ligand-binding properties and roles in immune response modulation.
