TIM50 also known as Translocase of Inner Mitochondrial Membrane 50 is a protein weighing approximately 50 kDa. This protein plays an important mechanical role in the mitochondrial presequence translocase functioning as a receptor and import channel within the inner mitochondrial membrane. TIM50 is expressed predominantly within the inner membranes of mitochondria in eukaryotic cells.
Biological function summary

TIM50 facilitates the import of presequence-containing proteins from the cytosol into the mitochondrial matrix acting as part of the TIM23 complex. This complex orchestrates the translocation of proteins across the inner mitochondrial membrane. By interacting with presequences of nuclear-encoded mitochondrial proteins TIM50 ensures proper import and processing which is important for mitochondrial function and biogenesis.

Pathways

TIM50 is critical in the mitochondrial protein import pathway which is essential for mitochondrial respiration and energy production. TIM50 interacts with other proteins such as TIM23 and TIM17 to form the translocase of the inner membrane 23 (TIM23) complex. TIM50 also links to the mitochondrial unfolded protein response (UPR) where it may influence the regulation of proteostasis within mitochondria alongside proteins like HSP70.

Mutations and dysregulation of TIM50 have connections to mitochondrial diseases which encompass a range of disorders due to mitochondrial dysfunction. Additionally TIM50 may affect neurodegenerative diseases where mitochondrial impairment plays a role. Alterations in TIM50 influencing mitochondrial function might correlate with proteins like cytochrome c which can contribute to apoptotic pathways and cell death in these diseases.
