Probable mitochondrial intermembrane chaperone that participates in the import and insertion of some multi-pass transmembrane proteins into the mitochondrial inner membrane. Also required for the transfer of beta-barrel precursors from the TOM complex to the sorting and assembly machinery (SAM complex) of the outer membrane. TIMM8B also known as Deafness Dystonia Protein 2 (DDP2) is a small protein with a molecular mass of about 10 kDa. It is expressed mainly in the mitochondria and is part of the intermembrane space machinery. TIMM8B plays a mechanical role in the mitochondrial protein translocase system helping transport specific proteins from the cytosol into the mitochondrial intermembrane space. The protein is part of a family that includes TIMM8A and it operates as a subunit of a larger assembly that aids in correct protein localization within the mitochondria.
TIMM8B contributes to cellular energy regulation by ensuring the proper functioning of the mitochondrial protein import machinery. TIMM8B forms a complex with TIMM13 another mitochondrial intermembrane space assembly. This complex assists in the import and insertion of essential proteins across the mitochondrial membranes which is important for mitochondrial respiratory chain and energy metabolism. Disruptions in this complex can impair mitochondrial function and cellular energy dynamics.
The mitochondria rely heavily on the TIMM8B-TIMM13 complex during the translocation process which influences the oxidative phosphorylation pathway. The complex components coordinate with the mitochondrial inner membrane translocase TIMM23 to ensure efficient energy production. Furthermore TIMM8B interacts indirectly with proteins such as cytochrome c as part of the electron transport chain highlighting its role in maintaining mitochondrial integrity and activity.
TIMM8B mutations or dysfunctions have been associated with auditory and neurological conditions such as Mohr-Tranebjaerg syndrome and possibly Charcot-Marie-Tooth disease. These diseases illustrate the protein's significance in sensory and nervous system function. TIMM8B may influence these diseases through its association with TIMM8A which has well-documented connections to similar auditory and neurological impairments.
