IHC image of ab28289 staining in human normal pancreas formalin fixed paraffin embedded tissue section, performed on a Leica BondTM system using the standard protocol F. The section was pre-treated using heat mediated antigen retrieval with EDTA (pH9, epitope retrieval solution 2) for 20 mins. The section was then incubated with ab28289, 1μg/ml, for 15 mins at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.

For other IHC staining systems (automated and non-automated) customers should optimize variable parameters such as antigen retrieval conditions, primary antibody concentration and antibody incubation times.

Tissue kallikrein also known as KLK1 is a serine protease enzyme that plays an important role in the cleavage of kininogen to produce kinins. This enzyme with a molecular weight of approximately 30 kDa exists in various tissues like the kidney pancreas and salivary glands. The protein is part of the kallikrein family and is expressed prominently in these areas. You can commonly measure its levels through ELISA assays labeled "kallikrein ELISA" or specific assays like the "kallikrein 3 ELISA."
Biological function summary

Tissue kallikrein contributes to vasodilation and blood pressure regulation by producing bradykinin a peptide with strong vasodilator properties. It does not function alone but may interact with other enzymes to amplify its effect.
