The TKTL1 protein also known as transketolase-like 1 or TKR is an important enzyme involved in cellular metabolism. It has a molecular mass of approximately 65 kDa. TKTL1 is widely expressed throughout various tissues with notably high levels of expression in the liver and brain. Mechanically it functions by participating in the non-oxidative branch of the pentose phosphate pathway facilitating the transfer of two-carbon units between sugar phosphates.
Biological function summary

TKTL1 supports numerous cellular processes by influencing the cell's redox state and nucleotide synthesis. Its activities help drive cell proliferation and survival often seen in rapidly dividing cells. Although not typically part of a macromolecular complex TKTL1 associates with other cellular components to execute its function. It plays a role in energy production and biosynthetic processes indirectly supporting the metabolic demands of cells.

Pathways

TKTL1 is involved in the pentose phosphate pathway and glycolysis. These pathways are critical for maintaining cellular homeostasis and generating the necessary biomolecules for cell growth. It operates alongside other proteins such as transketolase and transaldolase to ensure the proper flow of metabolites within these pathways. Additionally TKTL1's role affects the balance of nicotinamide adenine dinucleotide phosphate (NADPH) which is essential for reductive biosynthesis and antioxidant defense.

TKTL1 has associations with several cancers including colorectal and breast cancer where its expression levels can impact tumor growth and metastasis. The protein's connection to cancer is partly due to its influence on cell proliferation and metabolic reprogramming.
