Immunoprecipitation - Anti-TLR2 antibody [EPR20303] - Low endotoxin, Azide free (AB223545)
This IP data was generated using the same anti-TLR2 antibody clone [EPR20303] in a different buffer formulation ( cat# ab209217).
TLR2 was immunoprecipitated from 0.35 mg of RAW 264.7 (Mouse macrophage cell line transformed with Abelson murine leukemia virus) whole cell lysate, treated with 1 μg/ml LPS for 6 hours, with ab209217 at 1/30 dilution.
Western blot was performed from the immunoprecipitate using ab209217 at 1/500 dilution.
VeriBlot for IP Detection Reagent (HRP) (ab131366), was used for detection at 1/1000 dilution.
Lane 1 : RAW 264.7 whole cell lysate treated with 1 μg/ml LPS for 6 hours, 10 μg (Input).
Lane 2 : ab209217 IP in RAW 264.7 whole cell lysate treated with 1 μg/ml LPS for 6 hours.
Lane 3 : Rabbit monoclonal IgG (ab172730) instead of ab209217 in RAW 264.7 whole cell lysate treated with 1 μg/ml LPS for 6 hours.
Blocking and dilution buffer and concentration : 5% NFDM/TBST.
Exposure time : 1 second.

Product details
ab223545 is the carrier-free version of ab209217.
Patented technology
Our RabMAb® technology is a patented hybridoma-based technology for making rabbit monoclonal antibodies. For details on our patents, please refer to RabMAb® patents.
Conjugation ready
Our carrier-free antibodies are typically supplied in a PBS-only formulation, purified and free of BSA, sodium azide and glycerol. This conjugation-ready format is designed for use with fluorochromes, metal isotopes, oligonucleotides, and enzymes, which makes them ideal for antibody labelling, functional and cell-based assays, flow-based assays (e.g. mass cytometry) and Multiplex Imaging applications.
Use our conjugation kits for antibody conjugates that are ready-to-use in as little as 20 minutes with 1 minute hands-on-time and 100% antibody recovery: available for fluorescent dyes, HRP, biotin and gold.
Compatibility
This product is compatible with the Maxpar® Antibody Labeling Kit from Fluidigm, without the need for antibody preparation. Maxpar® is a trademark of Fluidigm Canada Inc.
What does low endotoxin mean?
Our low endotoxin, azide-free formats have low endotoxin level (1 EU/mg, determined by the TAL assay) and are free from azide, to achieve consistent experimental results in functional assays.

Biological function summary
TLR2 initiates signaling pathways that lead to the production of cytokines and chemokines helping to control infection. It forms dimers with other TLRs like TLR1 or TLR6 to expand its recognition range of microbial components. TLR2's activity is important for balancing pathogen recognition and avoiding overactive immune response. On macrophages TLR2 activation enhances their phagocytic ability.
Pathways
TLR2 plays roles in innate immune signaling pathways and inflammation pathways. It contributes significantly to the NF-kB and MAPK pathways which regulate inflammation and immune response. TLR2 often cooperates with proteins like MyD88 and TRAF6 in these pathways leading to downstream activation of transcription factors and production of pro-inflammatory molecules.
