Heat mediated antigen retrieval was performed using Tris/EDTA buffer pH 9. A prediluted HRP-polymer conjugated anti-rabbit IgG was used as the secondary antibody. Counterstained with hematoxylin.

TLR7 also known as Toll-like receptor 7 plays a mechanical role in the immune system by recognizing single-stranded RNA molecules. This receptor with a mass of approximately 120 kDa is a member of the Toll-like receptor family. TLR7 mainly expresses in immune cells like plasmacytoid dendritic cells and B cells. Its location on the endosomal surface allows it to detect viral RNA and trigger downstream signaling cascades.
Biological function summary

TLR7 induces an immune response by activating pathways that lead to the production of cytokines and type I interferons. It does not function alone but interacts within a complex involving other adaptors such as MyD88. This interaction initiates a signaling cascade that is essential for mounting an acute response to viral infections and boosting adaptive immunity.

Pathways

Single-stranded RNA detection through TLR7 activates the MyD88-dependent pathway which is important for innate immune responses. This pathway includes key components such as NF-κB and IRF7 leading to inflammatory cytokine production. TLR7 links with these proteins working together to activate the host defense mechanism against viral infections.

TLR7 plays a significant role in autoimmunity and viral infections. Aberrant activation of TLR7 has connections with systemic lupus erythematosus (SLE) where it may contribute to pathogenesis by promoting autoantibody production. Additionally TLR7 influences the immune response in chronic infections like hepatitis C where it operates with proteins like MyD88 and contributes to disease progression and severity.

Endosomal receptor that plays a key role in innate and adaptive immunity (PubMed : 14976261, PubMed : 32433612). Controls host immune response against pathogens through recognition of uridine-containing single strand RNAs (ssRNAs) of viral origin or guanosine analogs (PubMed : 12738885, PubMed : 27742543, PubMed : 31608988, PubMed : 32706371, PubMed : 35477763). Upon binding to agonists, undergoes dimerization that brings TIR domains from the two molecules into direct contact, leading to the recruitment of TIR-containing downstream adapter MYD88 through homotypic interaction (PubMed : 27742543). In turn, the Myddosome signaling complex is formed involving IRAK4, IRAK1, TRAF6, TRAF3 leading to activation of downstream transcription factors NF-kappa-B and IRF7 to induce pro-inflammatory cytokines and interferons, respectively (PubMed : 27742543, PubMed : 32706371).
